I de senere \u00e5r er forekomsten steget markant. Selvom medicinsk str\u00e5lebehandling\/kemoterapi og kirurgisk behandling har visse effekter p\u00e5 tidlig livmoderhalskr\u00e6ft, er behandlingseffekten p\u00e5 mellemliggende og sen livmoderhalskr\u00e6ft ikke god, og tilbagefaldsraten er h\u00f8j med \u00e5benlyse bivirkninger. E6\/E7, som de vigtigste onkogener i livmoderhalskr\u00e6ft, er \u00e5rsagen til indledende \u00e6ndringer i epitelceller og kan fremme v\u00e6rtscelleproliferation og viral amplifikation. Cinnamaldehyd (CA), som er hovedkomponenten i kanelens flygtige olie, er en naturlig lille molekyleforbindelse. Forskning har vist, at det har forskellige farmakologiske aktiviteter s\u00e5som feberneds\u00e6ttende, antiinflammatoriske og hypoglyk\u00e6miske virkninger og kan h\u00e6mme spredningen af forskellige tumorceller s\u00e5som ikke-sm\u00e5cellet lungekr\u00e6ft, leverkr\u00e6ft, mavekr\u00e6ft og melanom, hvilket udviser visse antitumoreffekter. Den molekyl\u00e6re mekanisme, der inducerer tumorcelleapoptose, er dog ikke blevet fuldt ud belyst, og der er kun f\u00e5 rapporter om dens virkninger mod livmoderhalskr\u00e6ft. Denne unders\u00f8gelse brugte Siha-celler fra livmoderhalskr\u00e6ft til at unders\u00f8ge CA's anticancer-effekt og mekanisme in vitro, hvilket giver yderligere teoretisk og eksperimentelt grundlag for forskning i livmoderhalskr\u00e6ft.<\/p>\n<p>&nbsp;<\/p>\n<p>Livmoderhalskr\u00e6ft er en af de mest almindelige gyn\u00e6kologiske maligniteter, ofte ledsaget af h\u00f8je tilbagefalds- og metastasefrekvenser, hvilket udg\u00f8r en alvorlig trussel mod kvinders sundhed. I de senere \u00e5r er naturlige forbindelser blevet udforsket som potentielle behandlinger af livmoderhalskr\u00e6ft ved at h\u00e6mme E6\/E7. For eksempel h\u00e6mmer tanshinon IIA E6\/E7 p\u00e5 transkriptionsniveau, hvilket f\u00f8rer til p53-afh\u00e6ngig v\u00e6ksth\u00e6mning i livmoderhalskr\u00e6ftceller; anisolinsyre nedregulerer E6\/E7-ekspression p\u00e5 proteinniveau i livmoderhalskr\u00e6ftceller gennem proteasomal nedbrydning og inducerer p53-uafh\u00e6ngig mitokondriel apoptose; N-benzylcinnamamid fremmer apoptose i humane livmoderhalskr\u00e6ftceller ved at h\u00e6mme E6\/E7 mRNA-niveauer. I denne unders\u00f8gelse rapporterede vi for f\u00f8rste gang, at CA kan fremkalde apoptose i Siha-celler og nedregulere ekspressionsniveauerne af HPV E6\/E7-proteiner.<\/p>\n<p>Unormal cellecyklus er en af hoved\u00e5rsagerne til tumordannelse, og dens reguleringsmekanismer omfatter cellecyklusdrift og overv\u00e5gning. N\u00e5r drivmekanismen er beskadiget, kan det f\u00f8re til ukontrolleret v\u00e6kst af normale celler og deres omdannelse til tumorceller; N\u00e5r overv\u00e5gningsmekanismen er beskadiget, kan det fremkalde funktionsforstyrrelser i DNA-reparation og celled\u00f8d, hvilket forv\u00e6rrer hele reguleringsmekanismen. Vi fandt gennem flowcytometri, at CA kan fremkalde en betydelig stigning i andelen af G2\/M-faseceller i Siha-celler, hvilket indikerer, at CA er blokeret i G2\/M-fasen. Apoptose er en form for programmeret celled\u00f8d, som hovedsageligt involverer d\u00f8dsreceptorens signalvej, mitokondriel signalvej og endoplasmatisk retikulums signalvej. De repr\u00e6sentative proteiner i den \"mitokondrielle signalvej\" er Bcl-2 og Bax, som aktiveres ved modtagelse af intracellul\u00e6re d\u00f8dssignaler og interagerer med hinanden i den ydre membran eller cytoplasma i mitokondrier, hvilket for\u00e5rsager \u00e6ndringer i mitokondriemembranens permeabilitet, tab af transmembranpotentiale og frigivelse af cytokrom C og andre proteiner. I det indledende eksperiment bekr\u00e6ftede vi f\u00f8rst gennem Hoechst 33342-farvning, at CA inducerer apoptose i Siha-celler; Derefter blev det gennem JC-1 fluorescerende sondep\u00e5visning fundet, at CA signifikant reducerede mitokondriernes transmembranpotentiale i Siha-celler; Yderligere Western blot-verifikation afsl\u00f8rede, at CA reducerede de anti-apoptotiske proteiner Bcl-2 og Survivin i Siha-celler, mens det \u00f8gede det pro-apoptotiske protein Bax, hvilket yderligere indikerer forekomsten af celleapoptose. E6\/E7-proteiner hj\u00e6lper med at opretholde den ondartede f\u00e6notype af livmoderhalskr\u00e6ftceller med HPV-infektion og kan regulere mange molekyler, der er relateret til celleoverlevelse og apoptoseveje. Denne artikel viser, at CA kan nedregulere ekspressionsniveauet af E6\/E7 og derved fremkalde apoptose i Siha-celler, der er inficeret med HPV. Efterf\u00f8lgende unders\u00f8gte vi den specifikke mekanisme for CA-induceret nedbrydning af E6\/E7. Resultaterne viste, at proteasom-inhibitoren MG132 reducerede den CA-inducerede E6\/E7-nedbrydning betydeligt, hvilket indikerer, at disse to proteiner tidligere blev nedbrudt af proteasomet.<\/p>\n<p>Virkningerne af sm\u00e5molekyl\u00e6re l\u00e6gemidler p\u00e5 genfunktionen er koncentrationsafh\u00e6ngige. Hvis et sm\u00e5molekyl\u00e6rt l\u00e6gemiddel kan ud\u00f8ve anti-tumorvirkninger, kan det bruge subletale koncentrationer til at screene for l\u00e6gemidler, der kan dr\u00e6be det synergistisk. Denne metode kaldes kemisk genomisk screeningsstrategi. P\u00e5 grund af selektiviteten af kemoterapeutiske l\u00e6gemidler til behandling af livmoderhalskr\u00e6ft i klinisk praksis og potentialet for l\u00e6gemiddelresistens og andre bivirkninger ved langvarig brug, er eksisterende kliniske kombinationsbehandlinger kun effektive for nogle patienter. Derfor er det forskningsarbejde, vi er n\u00f8dt til at udf\u00f8re, at finde og udvikle nye terapeutiske l\u00e6gemidler og kombinationsbehandlinger. I denne unders\u00f8gelse anvendte vi en kemisk genomisk screeningsstrategi til at p\u00e5vise den synergistiske effekt af CA med almindeligt anvendte livmoderhalskr\u00e6ftl\u00e6gemidler TAX, Pt og 5-FU i Siha-celler. Resultaterne bekr\u00e6ftede, at kombinationen af CA og de ovenn\u00e6vnte kemoterapimidler kan udvise synergistiske effekter ved en bestemt koncentration, og effekten af kombinationsbehandlingsgruppen er bedre end effekten af monoterapigruppen med statistisk signifikans. If\u00f8lge litteraturrapporter kan de ovenn\u00e6vnte kemoterapimedicin for\u00e5rsage DNA-skader og blokere cellecyklussen under antitumorprocessen; Blandt dem kan TAX blokere G2M-fasen; Pt kan blokere G2M-fasen; 5-FU kan blokere G0\/G1-fasen. If\u00f8lge forskningsresultaterne i denne artikel tyder det p\u00e5, at CA kan ud\u00f8ve en synergistisk effekt med de ovenn\u00e6vnte kemoterapimedicin ved i f\u00e6llesskab at blokere cellecyklussen og derved fremkalde celleapoptose og forbedre dens effekt mod livmoderhalskr\u00e6ft.<\/p>\n<p>Sammenfattende bekr\u00e6fter denne unders\u00f8gelse, at CA inducerer apoptose i Siha-celler gennem den mitokondrielle vej og nedregulerer udtrykket af HPV E6\/E7-proteiner gennem proteasomvejen; Samtidig kan det udvise synergistiske virkninger med forskellige almindeligt anvendte kemoterapimedicin til livmoderhalskr\u00e6ft p\u00e5 celleniveau.
