Fra et farmakologisk perspektiv er polyketidforbindelser en vigtig kilde til nye l\u00e6gemidler, der hovedsageligt bruges som antibiotika, immunosuppressiva, antiparasit\u00e6re midler, kolesterols\u00e6nkende midler og antitumorl\u00e6gemidler. Funktionen af hvert dom\u00e6ne i polyketidsyntase svarer til strukturen af det endelige produkt. Derfor kan denne korrespondance bruges til at redigere biosyntetiske gener og designe syntetiske produkter og dermed skabe et nyt forskningsfelt - kombinatorisk biologi af polyketidsyntase. Forskere genererer hovedsageligt nye produkter ved at reducere, \u00f8ge eller udskifte moduler og strukturelle dom\u00e6ner i polyketidsyntase, f.eks. ved at modificere polyketidsyntase-genklyngen aveLAT-ACP i Streptomyces avermitilis for at opn\u00e5 en genetisk manipuleret stamme. To nye avermectinderivater, 25 methyl ivermectin og 25 ethyl ivermectin, blev isoleret fra stammens sekund\u00e6re metabolitter.<\/p>\n<p>I den tidligere forskning modificerede vores forskningsgruppe milF-genet, der koder for polyketidsyntase i Streptomyces bingchengensis BC-120-4, for at opn\u00e5 den genetisk manipulerede stamme Streptomyces bingchengensis BCJ60. Denne stamme kan ikke kode for C-5 ketoreduktase og blokerer dermed for syntesen af milbeimycin A3\/A4\/B2\/B3. Gennem m\u00e5lrettet genetisk modifikation kan den biosyntetiske genklynge underg\u00e5 \u00e6ndringer, som kan aktivere visse biosyntetiske veje for at opn\u00e5 nye metabolitter. Derfor fokuserer denne unders\u00f8gelse hovedsageligt p\u00e5 de sekund\u00e6re metabolitter i den genetisk manipulerede stamme Streptomyces bingchengensis BCJ60 for at opn\u00e5 meget effektive insekticidantibiotika med praktisk v\u00e6rdi.<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>Denne artikel unders\u00f8gte systematisk de sekund\u00e6re metabolitter i den genetisk manipulerede bakterie Streptomyces bingchengensis BCJ60 og opn\u00e5ede 11 forbindelser, blandt hvilke forbindelser 1 og 2 er to nye forbindelser. I tidligere unders\u00f8gelser har vores forskningsgruppe udf\u00f8rt indledende forskning i de sekund\u00e6re metabolitter af den genetisk manipulerede bakterie Streptomyces bingchengensis BCJ60 og opn\u00e5et 12 nye forbindelser, som alle er milberelliner. Dette indikerer, at den biosyntetiske genklynge gennemg\u00e5r \u00e6ndringer gennem m\u00e5lrettet genmodificering, hvilket aktiverer visse biosyntetiske veje og giver nye metabolitter, som hj\u00e6lper med at forst\u00e5 den biosyntetiske mekanisme for Mirella-forbindelser yderligere. Eksperimentet med insektdr\u00e6bende aktivitet af den nye forbindelse viste, at forbindelserne 1 og 2 udviste signifikant dr\u00e6bende aktivitet mod cinnobermider og fyrretr\u00e6snematoder, og der var ingen signifikant forskel i forhold til det kommercielle Mirbeimycin A3\/A4.
