Den vokser i regnskoven i v\u00e5de tropiske dale og p\u00e5 skr\u00e5ninger ved flodbredder i 520-800 meters h\u00f8jde. Moderne forskning har vist, at honningplanten Kashiwagi indeholder masser af kemiske komponenter som sesquiterpener, triterpener, diterpener og limonoider. Nogle forbindelser har gode farmakologiske aktiviteter som f.eks. antibakterielle, antitumor- og smertestillende virkninger. Proteintyrosinfosfataser (PTP'er) er en gruppe af enzymer, der katalyserer defosforyleringen af proteintyrosin. Proteintyrosinfosfatase 1B (PTP-1B) spiller en afg\u00f8rende rolle i reguleringen af insulinvirkningen ved at aktivere insulinreceptorer og downstream-substratproteiner gennem defosforylering. Derfor er PIP-1B et vigtigt negativt reguleringsprotein i insulinsignaltransduktionsvejen, og PTP-1B-h\u00e6mmeren er et vigtigt valg til udvikling af l\u00e6gemidler mod diabetes. Forskerteamet fandt tidligt ud af, at diterpenoidkomponenterne i det flergrenede Kashiwagi-tr\u00e6 fra Meliaceae-familien havde en h\u00e6mmende effekt p\u00e5 PTP-1B, men denne aktivitet er ikke blevet rapporteret i andre planter af samme sl\u00e6gt. For yderligere at udforske de aktive ingredienser i Kashiwagi brugte dette eksperiment derfor silikagels\u00f8jlekromatografi, semiforberedende kromatografi og forberedende kromatografi kombineret med kernemagnetisk resonansspektroskopi til at unders\u00f8ge ethylacetatekstraktionsstedet for Kashiwagi-rodekstrakt for at give et grundlag for at h\u00e6mme opdagelsen af aktive ingredienser i PTP-1B.<\/p>\n<p>&nbsp;<\/p>\n<p>I denne unders\u00f8gelse blev i alt 23 komponenter isoleret fra ethylacetatekstraktet af Kashiwagi-roden og screenet for PTP-1B-h\u00e6mmende aktivitet. Baseret p\u00e5 dette blev forholdet mellem struktur og aktivitet forel\u00f8bigt udforsket. Forbindelserne 3, 16 og 21 er alle af cleanan-typen, mens forbindelse 12 er af prenyluledeman-typen og har den bedste h\u00e6mmende effekt, hvilket indikerer, at diterpener af prenyluledeman-typen har st\u00e6rkere h\u00e6mmende aktivitet end diterpener af cleanan-typen; Forbindelserne 3 og 21 er begge i E-konfiguration, mens forbindelse 16 er i Z-konfiguration. Forbindelse 16 udviser st\u00e6rk h\u00e6mmende aktivitet, kun overg\u00e5et af forbindelse 12, hvilket indikerer, at diterpenoidforbindelser af Z-typen har st\u00e6rkere h\u00e6mmende aktivitet; Forbindelser 3 og 16 indeholder acetylgrupper i deres strukturer og udviser god h\u00e6mmende aktivitet.
