F) Nees er en plante i familien Menispermaceae, som h\u00f8rer til meridianerne i hjertet, lungerne, tyktarmen og bl\u00e6ren. Den har en bitter smag og en kold natur. Den har en varmeafledende og afgiftende virkning, k\u00f8ler blodet og fjerner karbunkler. Andrographolid (molekyl\u00e6r formel C20H30O5, struktur vist i figur 1) er en hvid, firkantet krystal og det vigtigste ekstrakt af l\u00e6geplanten Andrographis paniculata. Forskning har vist, at Chuanxinlian-lakton har en bred vifte af biologiske aktiviteter, s\u00e5som antiinfektion, hepatobeskyttende og koleretisk, anticancer, antiinflammatorisk, antibakteriel, antimalaria, immunmodulerende, antikardiovaskul\u00e6r og cerebrovaskul\u00e6r sygdom, antiviral osv. Det bruges i vid udstr\u00e6kning i klinisk praksis og har betydelige effekter. If\u00f8lge litteraturen isolerede Boorsmas team f\u00f8rst en farvel\u00f8s krystal fra Houttuynia cordata, som senere blev strukturelt identificeret af Gorters team som en lactonforbindelse og navngivet andrographolid. P\u00e5 grund af dets d\u00e5rlige vandopl\u00f8selighed og stabilitet in vivo er biotilg\u00e6ngeligheden af Chuanxinlian-lakton lav, hvilket begr\u00e6nser dets kliniske anvendelse. Denne artikel introducerer hovedsageligt syntesen og aktivitetsforskningen af Chuanxinlian-laktonderivater ud fra fire aspekter: antitumor, antiinflammatorisk, antibakteriel og antiviral.<\/p>\n<p>&nbsp;<\/p>\n<p>Chuanxinlian lacton-derivater har en bred vifte af farmakologiske aktiviteter, og forskellige forskergrupper er stadig forpligtet til at bruge dem som blyforbindelser til at syntetisere en r\u00e6kke nye Chuanxinlian lacton-derivater med bedre aktivitet. P\u00e5 nuv\u00e6rende tidspunkt fokuserer modifikationstyperne af Chuanxinlian-lacton hovedsageligt p\u00e5 esterificering, etherificering, oxidation, acylering, amidation, dobbeltbindingsaddition, oxidation, Michael-additionsreaktion og intramolekyl\u00e6r cykliseringsreaktion af hydroxylgrupper. De nyligt syntetiserede derivatmolekyler udviser relativt st\u00e6rke antiinflammatoriske, antitumor-, antibakterielle og antivirale aktiviteter. Blandt dem forventes forbindelse 69 at blive et kandidatl\u00e6gemiddel til behandling af inflammation p\u00e5 grund af dets gode vandopl\u00f8selighed og lave cytotoksicitet, og forbindelse 94a forventes at blive udviklet som et nyt antibakterielt l\u00e6gemiddel. Virkningsmekanismen for derivatmolekyler er dog stadig stort set uklar, s\u00e5 de fleste aktive derivater anvendes stadig godt i klinisk praksis. I det fremtidige arbejde med derivatsyntese og aktivitetsscreening b\u00f8r der derfor anvendes computerst\u00f8ttet l\u00e6gemiddeldesign og andre metoder kombineret med aktivitetsverifikationseksperimenter for at unders\u00f8ge og udforske m\u00e5lene for aktive derivatmolekyler, styre den strukturelle modifikation af forbindelser, s\u00e5 flere derivatmolekyler kan komme ind i klinisk forskning som l\u00e6gemiddelkandidater og endda udvikle nye l\u00e6gemidler til klinisk anvendelse. Sammenfattende er forskere fra forskellige lande i \u00e5renes l\u00f8b aldrig holdt op med at studere derivater af paeoniflorin. Baseret p\u00e5 tidligere forskningsresultater s\u00f8ger de konstant nye veje til at syntetisere nye paeoniflorinderivater.
