Med den kraftige udvikling af dyrehold stiger menneskers eftersp\u00f8rgsel efter antibiotika ogs\u00e5 dag for dag. P\u00e5 nuv\u00e6rende tidspunkt er den \u00e5rlige produktion og brug af antibiotika i Kina ca. 189.000 tons, hvoraf en stor del anvendes i dyrehold. Brugen af veterin\u00e6re antibiotika udg\u00f8r mere end halvdelen af den samlede m\u00e6ngde antibiotika, der bruges i dyrehold. I 2010 overgik Kina Japan og USA med hensyn til animalske l\u00e6gemiddelr\u00e5varer og brug og blev verdens st\u00f8rste bruger af animalske l\u00e6gemiddelr\u00e5varer. Men den omfattende brug af antibiotika kan have alvorlige konsekvenser. Efter at v\u00e6re kommet ind i organismen absorberes nogle veterin\u00e6re antibiotika, mens andre overskrider det behandlede husdyrs toleranceomr\u00e5de og udskilles fra kroppen. Ved hj\u00e6lp af migration overf\u00f8res antibiotika til forskellige \u00f8kosystemer og ophobes l\u00f8bende i vand og jord, hvilket kan p\u00e5virke den normale funktion af organismer i milj\u00f8et. Derfor er effektiv og enkel behandling af restantibiotika i det nuv\u00e6rende milj\u00f8 for at forhindre yderligere skade p\u00e5 organismer blevet et varmt forskningsemne.<\/p>\n<p>P\u00e5 nuv\u00e6rende tidspunkt omfatter de vigtigste metoder til behandling af antibiotika fysisk adsorption, avanceret oxidation og mikrobiel nedbrydning. I dette eksperiment blev avilamycin derfor brugt som et repr\u00e6sentativt antibiotikum. H\u00f8jtydende v\u00e6skekromatografi blev brugt til at m\u00e5le nedbrydningsegenskaberne for Bacillus subtilis og Shigella p\u00e5 avilamycin under forskellige betingelser for temperatur, pH, pr\u00f8vevolumen, stammev\u00e6skevolumen og dyrkningstid. Response surface methodology blev brugt til at analysere nedbrydningsegenskaberne og opn\u00e5 de optimale betingelser for den mikrobielle stamme til at nedbryde avilamycin for at give et grundlag for den praktiske anvendelse af mikrobiel nedbrydning af avilamycin og andre veterin\u00e6re antibiotika.<\/p>\n<p>&nbsp;<\/p>\n<p>I de senere \u00e5r, med den gradvise udvikling af den gr\u00f8nne industri, er brugen af mikrobielle stammer til nedbrydning i behandlingen af milj\u00f8forurenende stoffer gradvist blevet et foretrukket emne for forskere. P\u00e5 grund af avermectins d\u00e5rlige vandopl\u00f8selighed er dets nedbrydningscyklus i mikrobielle nedbrydningsmetoder l\u00e6ngere end for tetracyklin og \u03b2-lactamantibiotika, og det tager normalt mere end 15 dage at opn\u00e5 en nedbrydningseffekt p\u00e5 over 80%. Forskning har vist, at kun et lille antal stammer kan nedbryde avermectin mere effektivt. For eksempel kan Burkholderia nedbryde ca. 80% avermectin inden for 48 timer, og termofile Bacillus kan opn\u00e5 en nedbrydningshastighed p\u00e5 77,6% avermectinstandard inden for 72 timer. Derfor er unders\u00f8gelse og screening af specifikke bakteriestammer gavnlig for den videre udvikling af mikrobielle nedbrydningsmetoder.<\/p>\n<p>Bakterier nedbryder generelt makrolidantibiotika ved at metabolisere dem og producere inaktiverende transferaser. Disse inaktiverende transferaser kan hovedsageligt opdeles i makrolidesteraser, 2'-glycosylphosphorylat transferaser MPH1 og glycosylphosphotransferaser, som kan fremme phosphorylering og glycosylering af antibiotiske molekyl\u00e6re strukturer og derved f\u00e5 antibiotika til at miste deres oprindelige egenskaber. Nedbrydningsmekanismen for stammer, der ofte bruges til antibiotikabehandling, er dog ikke blevet udforsket fuldt ud, hvilket ogs\u00e5 er et vanskeligt problem, som forskere skal l\u00f8se i dag. Sammenlignet med Bacillus subtilis har Shigella dog en lavere optimal nedbrydningstemperatur, hvilket er gavnligt for at reducere energiforbruget under den faktiske nedbrydning.
