Folk er i stigende grad bekymrede over forskning og udvikling af nye alternative energikilder, og cellulosebiomassebr\u00e6ndstof er i \u00f8jeblikket anerkendt som den mest v\u00e6rdifulde og lovende vedvarende energikilde. Cellulosebiomasse er den mest udbredte og rigelige biomasse p\u00e5 jorden og best\u00e5r hovedsageligt af cellulose, hemicellulose og lignin. Cellulose, hemicellulose og lignin er dog t\u00e6t bundet gennem hydrogen- og kovalente bindinger og har en st\u00e6dig biomassestruktur. Vi har kun udviklet mindre end 2% celluloseressourcer. Brugen af moderne bioteknologi til at omdanne cellulose til flydende br\u00e6ndstoffer som f.eks. ethanol har et stabilt, vedvarende og forureningsfrit udbytte og har potentiale til at l\u00f8se problemer som energikrise, milj\u00f8forurening og f\u00f8devarekrise. Men for at l\u00f8se dette problem er det mest afg\u00f8rende at f\u00e5 effektive og billige cellulaser.<\/p>\n<p>Cellulase er en f\u00e6llesbetegnelse for en gruppe komplekse enzymsystemer, der nedbryder cellulose til monosakkarider eller polysakkarider, prim\u00e6rt endoglucanase, exoglucanase og \u03b2-1,4-glucosidase. Disse tre enzymer arbejder sammen om at nedbryde cellulosestoffer. P\u00e5 nuv\u00e6rende tidspunkt fokuserer forskningen i cellulaseproducerende bakterier hovedsageligt p\u00e5 svampe som Trichoderma, Penicillium og Aspergillus. Men de fleste af deres cellulaser binder sig til cellemembranen, hvilket er besv\u00e6rligt at bruge, og omkostningerne ved enzymseparation og -ekstraktion er for h\u00f8je. Bakterier vokser hurtigere end svampe, kan opn\u00e5 h\u00f8jere ekspressionsniveauer af rekombinante enzymer og kan producere mere komplekse glycosidhydrolaser til synergistiske reaktioner med enzymatisk hydrolyse. I mellemtiden har bakterier en h\u00f8j naturlig diversitet og er mere tilb\u00f8jelige til at producere varmebestandige og alkalistabile enzymer. Indtil videre omfatter de cellulaseproducerende bakterier, der er blevet grundigt unders\u00f8gt, Vibrio, Bacillus, Bacteroides, Clostridium, Erwinia, Ruminococcus og Thermomonas.<\/p>\n<p>Selv om der hidtil er blevet forsket meget i omdannelsen af lignocellulose, st\u00e5r omdannelsen af cellulosebiomasse til bioethanol stadig over for store udfordringer p\u00e5 grund af de h\u00f8je kommercielle omkostninger ved hemicellulaser og cellulaser. Derfor haster det med at udvikle nye, effektive og billige cellulaser, og opdr\u00e6t af h\u00f8jtydende cellulasestammer er s\u00e6rligt vigtigt. Screening af h\u00f8jtydende cellulaseproducerende bakteriestammer fra laboratoriebevarede stammer, udf\u00f8relse af identifikation og forskning i dem, tilvejebringelse af stammekilder til forskning i cellulasepr\u00e6paration og l\u00e6gning af et vist fundament for genetisk forbedring, genteknologisk stammekonstruktion og andet arbejde.<\/p>\n<p>&nbsp;<\/p>\n<p>I denne unders\u00f8gelse blev i alt 5 bakteriestammer med h\u00f8j cellulaseproduktion screenet fra laboratoriekonserverede stammer. Efter morfologiske, fysiologiske og biokemiske egenskaber samt 16S rDNA-analyse blev de identificeret som Bacillus subtilis, 2 stammer af Bacillus subtilis og 3 stammer af Bacillus amyloliquefaciens. Mange svampe og bakterier i naturen kan producere forskellige cellulaser. P\u00e5 nuv\u00e6rende tidspunkt er der mange unders\u00f8gelser af svampecellulaser, og cellulaserne p\u00e5 markedet kommer hovedsageligt fra svampe. De cellulaseproducerende bakterier har ogs\u00e5 tiltrukket sig stor opm\u00e6rksomhed p\u00e5 grund af deres st\u00e6rke tilpasningsevne og andre fordele, og de vil sandsynligvis blive en meget vigtig kilde til cellulase i industrien. Blandt dem kan Bacillus subtilis producere og udskille et stort antal ekstracellul\u00e6re enzymer, hvilket g\u00f8r den til den vigtigste studerede bakterie. Derudover kan de ogs\u00e5 danne endosporer og sekund\u00e6re metabolitter under forhold med langsom v\u00e6kst i cellulosematrix, hvilket giver dem en konkurrencem\u00e6ssig fordel. Blandt dem er Bacillus subtilis den mest rapporterede fremragende cellulaseproducerende bakterie, mens Bacillus amyloliquefaciens sj\u00e6ldent er rapporteret.<\/p>\n<p>Forholdet mellem hydrolysecirkeldiameteren og kolonidiameteren p\u00e5 Congo-r\u00f8d CMC-pladen i denne artikel er st\u00f8rre end 4,5, hvor den mindste ZB6 er 4,61 og den maksimale AF1-bakterieforhold n\u00e5r 8,04, hvilket langt overstiger de bakteriestammer med h\u00f8j enzymaktivitet, der er screenet ved hj\u00e6lp af den samme metode som rapporteret i eksisterende indenlandsk og udenlandsk litteratur. Den h\u00f8jeste enzymaktivitet, der er rapporteret i litteraturen, var en stamme PX19, der blev isoleret af Tang Hao et al. fra tarmkanalen hos den gigantiske bambuselefant. Forholdet mellem hydrolysecirkeldiameter og kolonidiameter for stammen med den h\u00f8jeste enzymaktivitet var kun 4,83. Efter optimering og screening af de faste fermenteringsbetingelser for cellulaseproduktion af AF1-stammen kan dens h\u00f8jeste filterpapirenzymaktivitet n\u00e5 26,904U\/g. Derudover fandt det tidligere forskerhold, at AF1-bakterier ogs\u00e5 kan producere meget aktiv amylase, protease og bredspektrede antibakterielle stoffer. N\u00e5r den bruges til fodring af kyllinger, kan den forbedre foderudnyttelsen betydeligt, h\u00e6mme patogenernes v\u00e6kst, reducere sygdomsforekomsten og s\u00e6nke d\u00f8deligheden.
