Lakrids udviser fremragende anti CRPC-aktivitet, og glycyrrhizin i lakrids er en af de mange flavonoider, der har tiltrukket sig stor opm\u00e6rksomhed og har god anti-tumoraktivitet. For bedre at forst\u00e5 glycyrrhizins virkningsmekanisme p\u00e5 CRPC brugte vi netv\u00e6rksfarmakologiske metoder til at indsamle 683 m\u00e5l for glycyrrhizin gennem databaser som TCMIO, TCMSP og PharmMapper. Vi fandt 55 vigtige m\u00e5l for glycyrrhizin mod kastrationsresistent prostatakr\u00e6ft ved hj\u00e6lp af Venn-diagramv\u00e6rkt\u00f8jer og identificerede 10 kernem\u00e5l som AKT1, TP53, ESR1, EGFR og ALB gennem PPI-proteininteraktionsnetv\u00e6rksanalyse. GO-berigelsesanalyse viste, at glycyrrhizin hovedsageligt regulerer cellemetabolisme og apoptose ved at regulere proteinphosphorylering, oxidationsreaktioner og cellecyklus. Molekyl\u00e6r docking viste endvidere, at glycyrrhizin har god dockingaktivitet med 10 kernem\u00e5l. Det forudsiges, at glycyrrhizin kan opn\u00e5 antitumoreffekter ved at regulere udtrykket af disse kernem\u00e5lproteiner. Blandt disse kernem\u00e5l har AKT1-proteinet den h\u00f8jeste dockingaktivitet. AKT1 er en af de vigtige regulerende faktorer i PI3K\/AKT-signalvejen, der er involveret i forskellige biologiske processer i tumorer. I prostatakr\u00e6ft blev genetiske variationer og dysregulering af genekspression af PI3K\/AKT-signalvejskomponenter p\u00e5vist i op til 42% af prim\u00e6r prostatakr\u00e6ft og 100% af metastatiske prostatakr\u00e6ftpr\u00f8ver baseret p\u00e5 genom- og transkriptomprofiler. I tidlige unders\u00f8gelser blev det foresl\u00e5et, at AKT-undertyper (AKT1, AKT2) kan fremme overgangen fra androgenf\u00f8lsomt stadium til hormonrefrakt\u00e6rt stadium i prostatakr\u00e6ft. P\u00e5 baggrund af grundforskning i behandling af CRPC-patienter har unders\u00f8gelser rapporteret, at knockout af AKT1\/2 i kastrationsresistente prostatacancer-dyremodeller har reduceret in vivo- og in vitro-metastaser betydeligt. Det kan ses, at h\u00e6mning af ekspressionen af AKT1 kan forbedre overlevelsesprognosen for CRPC-patienter, og AKT1-h\u00e6mmere kan v\u00e6re en potentiel behandling for CRPC-patienter. Vores unders\u00f8gelse antyder teoretisk, at glycyrrhizin kan v\u00e6re en AKT1-h\u00e6mmer, men om glycyrrhizin kan blive en effektiv AKT1-h\u00e6mmer for CRPC-patienter, skal stadig verificeres yderligere eksperimentelt i fremtiden.<br \/>\nResultaterne af KEGG-vejberigelsen viste, at PI3K\/AKT-signalvejen og proteoglykaner var involveret i 105 relaterede signalveje, herunder kr\u00e6ft, prostatakr\u00e6ft, kemokinreceptoraktivering, endokrin resistens og resistens over for epidermal v\u00e6kstfaktorreceptortyrosinkinaseh\u00e6mmere (P&lt;0,01). Blandt dem er phosphatidyl-3-kinase (PI3K)\/serin-threonin-proteinkinase (AKT)-signalvejen den mest bekymrende. PI3K\/AKT-signalvejen er involveret i regulering af cellul\u00e6r metabolisme, v\u00e6kst, proliferation, differentiering og proteinsyntese. Mutationer i PI3K\/AKT-signalvejen er blevet p\u00e5vist i mange humane tumorer. Forskningsdata viser, at op til 70% til 100% af patienter med fremskreden prostatakr\u00e6ft oplever dysregulering af PI3K-signalvejen. Og det blev konstateret, at PI3K\/AKT-signalvejen spiller en vigtig rolle i udviklingen af hormonf\u00f8lsom prostatacancer til kastrationsresistent prostatacancer, og dens virkningsmekanisme kan v\u00e6re relateret til reguleringen af androgenreceptorens (AR) signalvej, som kan tjene som en potentiel biomark\u00f8r for CRPC. P\u00e5 nuv\u00e6rende tidspunkt er inhibitorer relateret til PI3K\/AKT-signalvejen, s\u00e5som AZD5363, g\u00e5et ind i den kliniske fors\u00f8gsfase og har opn\u00e5et betydelige terapeutiske effekter. Unders\u00f8gelse af h\u00e6mmere relateret til denne vej har betydelig v\u00e6rdi i behandlingen af CRPC.<br \/>\nVi fandt desuden ved hj\u00e6lp af CCK8-metoden, at glycyrrhizin kan h\u00e6mme spredningen af PC-3-celler betydeligt. Flowcytometri og DAPI-farvningseksperimenter viste, at det kan inducere celleapoptose, og et fald i celleantal og morfologisk krympning blev observeret under mikroskopet. Til screening af antitumorl\u00e6gemiddelkoncentrationer fandt vi, at deres IC50 var lidt h\u00f8jere, hvilket er relateret til den semifleksible dockingmetode, der blev brugt til molekyl\u00e6r docking. Sammenlignet med fleksibel docking er bindingsenergien beregnet ved semifleksibel docking lavere. Som en naturlig antioxidant har glycyrrhizin desuden st\u00e6rke antioxidanteffekter. Lavdosis naturlige antioxidanter viste imidlertid ikke god antitumoraktivitet i antitumoreksperimenter, mens h\u00f8jdosis naturlige antioxidanter har toksiske virkninger p\u00e5 celler, og dens specifikke mekanisme skal udforskes yderligere. Baseret p\u00e5 KEGG-signalvejen, der forudsiges af netv\u00e6rksfarmakologi, forudsiger vi, at den h\u00e6mmende effekt af glycyrrhizin p\u00e5 PC-3-celler hovedsageligt kan v\u00e6re relateret til PI3K\/AKT-signalvejen. Yderligere Western blot-eksperimenter blev udf\u00f8rt for at unders\u00f8ge effekten af glycyrrhizin p\u00e5 ekspressionen af proteiner relateret til PI3K\/AKT-signalvejen i PC-3-celler. Resultaterne viste, at glycyrrhizin kan h\u00e6mme fosforyleringsprocessen af AKT-proteiner betydeligt. I vores eksperiment blev det observeret, at glycyrrhizin kan h\u00e6mme ekspressionen af fosforyleret AKT-protein betydeligt, men den h\u00e6mmende effekt p\u00e5 ekspressionen af fosforyleret PI3K-protein var ikke som forventet. Efter gentagne gange at have justeret proteinbelastningskoncentrationen og -forholdet og strengt standardiseret operationen, viste proteinet stadig ikke signifikante effekter. Det vurderes, at \u00e5rsagen kan v\u00e6re relateret til kvalitetsproblemet med selve p-PI3K-proteinet, som har lav styrke. P\u00e5 grund af begr\u00e6nsede midler til denne unders\u00f8gelse erstattede vi dog ikke proteinerne fra andre produkter. I fremtidige eksperimenter vil vi analysere flere besl\u00e6gtede proteiner fra denne vej for at verificere vores hypotese.<br \/>\nSammenfattende analyserede og forudsagde denne unders\u00f8gelse den potentielle virkningsmekanisme for glycyrrhizin p\u00e5 kastrationsresistent prostatakr\u00e6ft gennem netv\u00e6rksfarmakologi og verificerede den yderligere gennem molekyl\u00e6r docking og in vitro-celleeksperimenter. Resultaterne viste, at glycyrrhizin kan h\u00e6mme proliferationen og inducere apoptose af PC-3-celler, og dens mekanisme er relateret til fosforyleringsprocessen af AKT-protein. Derfor spekulerer vi i, at dets anti CRPC-mekanisme hovedsageligt kan v\u00e6re gennem regulering af fosforylering af proteiner relateret til PI3K\/AKT-signalvejen.
