Gammel litteratur fort\u00e6ller, at den har funktionerne \"at fordrive vind og fugt, afgifte og opl\u00f8se blodstase, reducere h\u00e6velse og lindre smerte\", hvilket er i tr\u00e5d med behandlingsprincippet \"at afgifte og fjerne blodstase, eliminere slim og opl\u00f8se knuder\" for lungekr\u00e6ft. Forskning har vist, at b\u00e5de den stinkende p\u00e6onforbindelse og dens ekstrakt har god antitumoraktivitet, is\u00e6r den bedste antitumoraktivitet mod lungekr\u00e6ft. I den indledende forskning gennemf\u00f8rte forskergruppen flere flydende kvalitetsanalyser af afkoget og forskellige ekstraherede komponenter af stinkende p\u00e6on og fandt, at hovedkomponenterne i stinkende p\u00e6on er phenylethanoliske glykosider, blandt hvilke hovedkomponenterne er bladlignende og heterogene bladlignende glykosider. Efter eksperimentel screening og demonstration blev det konstateret, at phenylethanoliske glykosider fra stinkende p\u00e6on har gode antitumoreffekter. Derfor spekuleres der i, at de phenylethanoliske glykosider, der hovedsageligt best\u00e5r af bladlignende og heterogene bladlignende glykosider, kan v\u00e6re hovedkomponenterne i den stinkende p\u00e6ons antitumoreffekt.<\/p>\n<p>EphA2 er det vigtigste medlem af Eph-familien, den st\u00f8rste underfamilie af receptortyrosinkinaser (RKT'er), og er overudtrykt i forskellige ondartede tumorer, is\u00e6r i tumorer med h\u00f8j malignitet og st\u00e6rk invasion og metastaseevne, og er involveret i invasionen og metastasen af lungekr\u00e6ft. Forskning har vist, at EphA2-receptorer udtrykkes signifikant hos lungekr\u00e6ftpatienter og lungekr\u00e6ftcellelinjer, og jo h\u00f8jere ekspressionsniveau, jo v\u00e6rre er patienternes prognose. Ikke-ligandafh\u00e6ngig EphA2-signalering kan fremme tumorproliferation og metastase, forbedre tumorl\u00e6gemiddelresistens og opretholde tumorstamcellelignende egenskaber. Det kan ses, at EphA2 har dobbelte aktiviteter af ligandaktiveret anticancer og ikke-ligandaktiveret pro-cancer, og dephosphorylering eller phosphorylering af EphA2 S897-stedet er den kontakt, der styrer de to modsatte biologiske effekter. Defosforyleringen af EphA2 S897 er relateret til liganden EphrinA1, mens fosforyleringen p\u00e5 EphA2 S897 er relateret til ATK, som er den tidligste og anerkendte vigtige faktor. I denne unders\u00f8gelse fandt vi, at ekstrakter af Paeonia lactiflora phenylethanoliske glycosider signifikant kan h\u00e6mme spredning, migration og invasion af A549-celler samt undertrykke ekspressionen af EphA2, mTOR, p-mTOR, \u03b2 - catenin, GSK-3 \u03b2, P-GSK-3 \u03b2 og P-ATK-proteiner. Blandt dem er nedregulering af P-ATK s\u00e6rlig vigtig, og nedregulering af EphA2-protein kan ogs\u00e5 opregulere udtrykket af EphrinA1. Det phenylethanoliske glycosidekstrakt af stinkende p\u00e6on kan inducere bindingen af EphA2 med ligand EphrinA1, fremme ligand EphrinA1-afh\u00e6ngig anti-cancer-vej og ud\u00f8ve antitumoreffekt ved at h\u00e6mme EphA2, P-ATK, mTOR og andre faktorer og blokere EphA2\/ATK\/mTOR-signalvejen. Stigningen i P-EphA2 skyldes ligandafh\u00e6ngighed. Selvom S897-stedet p\u00e5 EphA2 gennemg\u00e5r dephosphorylering, aktiveres EphA2-tyrosinkinase ved phosphorylering, s\u00e5 den samlede P-EphA2 kan blive opreguleret.
