De sekund\u00e6re metabolitter, der stammer fra actinomyceter, udg\u00f8r ca. 45% af de sekund\u00e6re metabolitter, der stammer fra mikroorganismer, og udviser en r\u00e6kke biologiske aktiviteter, herunder antitumor, antibakterielle, antiinflammatoriske, antivirale og antiangiogene virkninger. Antibiotika, der stammer fra actinomyceter, udg\u00f8r 70% af de rapporterede naturlige antibiotika, og nogle af dem er blevet brugt meget i klinisk praksis, f.eks. vancomycin og azithromycin. Actinobakterier har altid v\u00e6ret anset for at v\u00e6re en god kilde til nye aktive sekund\u00e6re metabolitter fra mikrobielle kilder.<br \/>\nPolaromr\u00e5der omfatter generelt Nord- og Sydpolen og deres underomr\u00e5der, der er kendetegnet ved lave temperaturer, polart dagslys og st\u00e6rk ultraviolet str\u00e5ling. Polare mikroorganismer har udviklet en unik genregulering og metaboliske funktioner for at tilpasse sig ekstreme polare forhold og har dermed potentiale til at producere strukturelt nye sekund\u00e6re metabolitter. I de senere \u00e5r har forskere opdaget mange strukturelt unikke og meget aktive sekund\u00e6re metabolitter fra pol\u00e6re actinomyceter. Denne artikel gennemg\u00e5r de forskningsm\u00e6ssige fremskridt inden for sekund\u00e6re metabolitter og farmakologiske aktiviteter fra pol\u00e6re actinomyceter fra 1999 til 2021. I henhold til deres kemiske strukturtyper kan de klassificeres i peptider, alkaloider, terpener, makrolider, polyketoner, makrolactamer og andre kategorier. Oversigten over nye sekund\u00e6re metabolitter er vist i tabel 1. De fleste af de sekund\u00e6re metabolitter fra pol\u00e6re actinomyceter blev isoleret fra Streptomyces-sl\u00e6gten (74,5%), mens en lille del blev isoleret fra pseudo-Nocardia-sl\u00e6gten (14,2%), termofile Bacillus-sl\u00e6gter (2,8%), Nocardia-lignende sl\u00e6gter (0,9%) og andre sl\u00e6gter (7,7%) (Figur 8, til venstre). If\u00f8lge klassificeringen af kemiske strukturtyper er 21 (20,2%) af de 104 sekund\u00e6re metabolitter peptidforbindelser, 9 (8,7%) er alkaloider, 11 (10,6%) er terpener, 15 (14,4%) er makrolider, 19 (18,3%) er polyketoner, 6 (5,8%) er makrolactamer, og 23 (22,1%) er andre typer af forbindelser. De vigtigste strukturelle typer er peptider, polyketoner, makrolider osv. (figur 8, h\u00f8jre). Blandt dem omfatter de sekund\u00e6re metabolitter med meget sj\u00e6ldne strukturer nitroporosiner A (29) og B (30), antartin (41), gephyromycin (41) osv. 60), saliniquinoner G-I (72-74) osv. De fleste af de 104 sekund\u00e6re metabolitter har gode farmakologiske aktiviteter som f.eks. antibakterielle, svampedr\u00e6bende, antivirale og cytotoksiske. Det er hovedsageligt kendetegnet ved antitumoraktivitet, som har h\u00e6mmende aktivitet p\u00e5 leuk\u00e6mi, tyktarmskr\u00e6ft, epidermal kr\u00e6ft, myelom, livmoderhalskr\u00e6ft, brystkr\u00e6ft, melanom, gastrisk kr\u00e6ft og andre tumorceller. Nogle forbindelser har signifikant aktivitet, s\u00e5som C-1027 chromophore-V har signifikant cytotoksicitet, saliniquinoner G \uff5e H har signifikant antibakteriel aktivitet, og N - (2-hydroxypheny1) -2-phenazinamin (NHP) har signifikant antifungal og cytotoksicitet. Derudover har nogle forbindelser ogs\u00e5 selektiv h\u00e6mmende effekt. Det n\u00e6ste er antibakteriel aktivitet, herunder antibakteriel og antifungal aktivitet, som har h\u00e6mmende virkninger p\u00e5 Candida albicans, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis, Proteus mirabilis, Bacillus cereus, Bacillus subtilis og Micrococcus luteus. Nogle f\u00e5 har effekter som antiangiogenese, antioxidant, reduktion af cellul\u00e6re lipidniveauer, h\u00e6mning af melaninsyntese og glutaminaktivitet. Yderligere forskning og udvikling er berettiget for sekund\u00e6re metabolitter med nye strukturer, selv sj\u00e6ldne skeletter, og betydelig aktivitet, s\u00e5som nitrosporosiner A (29) og B (30), antagonist (41), gephyromycin (60) og salinequine G-I (72-74).<br \/>\nSammenfattende har polare actinomyceter rigelige ressourcer, hvor Streptomyces er hovedfokus for forskningen. Andre sj\u00e6ldne arter har brede udviklingsmuligheder. De kemiske strukturtyper af sekund\u00e6re metabolitter produceret af pol\u00e6re actinomyceter er forskellige, og mange af dem har god biologisk aktivitet, is\u00e6r antitumoraktivitet, hvilket har stor v\u00e6rdi for l\u00e6gemiddeludvikling. Der er dog ikke blevet forsket meget i metabolitternes biosynteseveje, aktive mekanismer og kliniske aspekter, og de fleste af de sekund\u00e6re metabolitter, der er isoleret fra pol\u00e6re actinomyceter, er kendte forbindelser. Med udviklingen af bioinformatik-teknologi er flere genomsekvenser af pol\u00e6re actinomyceter blevet opn\u00e5et gennem sekventering, og flere og flere pol\u00e6re actinomycete BGC'er er blevet opdaget og rapporteret, s\u00e5som PKS, NRPS-gener, halogenerede enzymgener, terpen- og oligosaccharidderivatrelaterede gener osv. Man kan se, at pol\u00e6re actinomyceter har et st\u00e6rkt potentiale for at syntetisere aktive sekund\u00e6re metabolitter og tilpasse sig pol\u00e6re milj\u00f8er, hvilket vil give rigere og mere forskelligartede kilder til nye forbindelser til udvikling af nye l\u00e6gemidler. Samtidig vil de biosyntetiske veje for nye sekund\u00e6re metabolitter v\u00e6re lettere at dechifrere og er v\u00e6rdige til yderligere forskning og udvikling. Selvom de fleste af de stoffer, der er isoleret fra pol\u00e6re actinomyceter, i \u00f8jeblikket er kendte, er det ikke sikkert, at de \"tavse gener\", der er ansvarlige for biosyntesen af nye stoffer, kommer til udtryk under fermenteringen. Derfor kan udtrykket af \"tavse gener\" i stammerne stimuleres ved at optimere fermenteringsbetingelserne, mikrobiel samkultur og andre metoder. Kombineret med genmining, aktivitetsscreening og andre metoder kan forbindelsesisolering styres til at isolere mere aktive sekund\u00e6re metabolitter med nye strukturer fra pol\u00e6re actinomycete-ressourcer.
