P\u00e5 nuv\u00e6rende tidspunkt er avernectiner de mest anvendte antiparasit\u00e6re l\u00e6gemidler i veterin\u00e6r klinisk praksis verden over. Men p\u00e5 grund af misbrug af dette l\u00e6gemiddel er der opst\u00e5et l\u00e6gemiddelresistente stammer i mange omr\u00e5der. I slutningen af 1980'erne introducerede og udbredte Kina denne type l\u00e6gemiddel, og nu har gastrointestinale nematoder hos tamf\u00e5r udviklet varierende grader af resistens over for avermectin-l\u00e6gemidler. Startect\u00ae er en ny type oralt korttidsvirkende ormemiddel til f\u00e5r, der er udviklet af Zoetis, verdens st\u00f8rste l\u00e6gemiddel- og vaccinevirksomhed inden for dyresundhed, og som effektivt kan forebygge og l\u00f8se parasit\u00e6re sygdomme hos husdyr og fjerkr\u00e6 og deres resistensproblemer. De aktive ingredienser i dette l\u00e6gemiddel er derquantel (10mg\/mL) og avermectin (1mg\/mL). Standarddosis af Startect \u00ae er 1 ml\/5 kg (v\u00e6skevolumen\/f\u00e5rets kropsv\u00e6gt), og hvert 50 kg voksent f\u00e5r kr\u00e6ver ca. 0,1 g Dexmedetomidine, der skal administreres \u00e9n gang. If\u00f8lge statistikkerne fra FN's F\u00f8devare- og Landbrugsorganisation (FAO) er verdens f\u00e5rebestand n\u00e5et op p\u00e5 1,2 milliarder. Derfor er den potentielle eftersp\u00f8rgsel efter Dequnt i hundredvis af tons eller endnu h\u00f8jere, og markedseftersp\u00f8rgslen er enorm.<\/p>\n<p>Dextrant er et kemisk derivat af svampemetabolitten paraherquamid A (PA), som hovedsageligt opn\u00e5s gennem amidcarbonylreduktion og deoxygenering af PA (se figur 1). Derfor er den industrielle r\u00e5vareforsyning af Dequnte hovedsageligt p\u00e5virket af PA-produktion. Den kemiske struktur af naturlige produkter som paraherquamid er imidlertid kompleks. Selvom de kan syntetiseres kemisk fra industrielle r\u00e5materialer som isopren eller isoprenepoxider, kan deres kemiske reaktionstrin n\u00e5 op p\u00e5 46 trin, hvilket resulterer i h\u00f8je omkostninger og ekstremt lave udbytter (2,5%), hvilket g\u00f8r det vanskeligt at producere i industriel skala. Produktionen af PA gennem mikrobiel fermentering er i \u00f8jeblikket en effektiv m\u00e5de at opn\u00e5 denne forbindelse p\u00e5 i stor skala. PA blev f\u00f8rst isoleret fra fermenteringsproduktet fra en stamme af Penicillium paraherquei, en svamp, der tilh\u00f8rer sl\u00e6gten Penicillium. Styrken af PA produceret af vildtypen Penicillium var dog ikke h\u00f8j, hvilket forhindrede yderligere eksperimentel forskning og industriel anvendelse. I anvendelsesforskningen af svampemetabolitter afh\u00e6nger styrken af m\u00e5lmetabolitten ikke kun af selve stammens ydeevne, men endnu vigtigere af at optimere fermenterings- og dyrkningsbetingelserne for at udnytte dens produktionskapacitet fuldt ud. Responsoverflademetoden bruges i \u00f8jeblikket i vid udstr\u00e6kning til at optimere mikrobiel fermenteringskultur. Denne metode kombinerer eksperimentelt design og matematisk modellering og har fordelene ved f\u00e6rre eksperimenter, kortere cyklusser, h\u00f8jere n\u00f8jagtighed ved opn\u00e5else af regressionsligninger, bedre forudsigelighed og unders\u00f8gelse af samspillet mellem flere faktorer.<\/p>\n<p>I det indledende arbejde opdagede vores forskningsgruppe en marineafledt Penicillium-svamp KWF31 med h\u00f8j PA-produktion. Den indledende unders\u00f8gelse viste, at dens fermenteringspotentiale var 55,8 \u00b1 3,7 mg\/L under Chatelier-mediet, hvilket ikke opfyldte kravene til industriel produktion. Det er et r\u00e5materiale til syntese af det kommercielle insektresistente middel Dextrant og har en h\u00f8j markedsv\u00e6rdi og stor eftersp\u00f8rgsel p\u00e5 markedet. Den vilde Penicillium-stamme har dog et lavt udbytte af PA, med flere kemiske syntesetrin og lavt udbytte, hvilket g\u00f8r det vanskeligt at im\u00f8dekomme markedets eftersp\u00f8rgsel. Denne unders\u00f8gelse brugte Plackett Burman-design til at screene tre mediumkomponenter, der signifikant p\u00e5virker den horisontale fermenteringsproduktion af PA af vildstammen KWF31 i rystekolber fra otte faktorer: opl\u00f8selig stivelse, glukose og g\u00e6rekstrakt; Deres koncentrationer blev gradvist optimeret ved hj\u00e6lp af klatreeksperimenter og Box Behnken-design. De optimerede optimale koncentrationer er opl\u00f8selig stivelse 31,0 g\/l, glukose 1208 g\/l og g\u00e6rekstrakt 14,5 g\/l. I dette medium viste vildtypestammen KWF31 en h\u00f8j styrke p\u00e5 243,7 mg\/L for PA-produktion under horisontal fermentering i rystekolbe, hvilket var 3,4 gange h\u00f8jere end f\u00f8r optimeringen (55,8 mg\/L). Den proces, der blev etableret i denne unders\u00f8gelse til fermenteringsproduktion af PA af den marine svamp KWF31, har god reproducerbarhed, stabilitet og kontrollerbarhed.
