Derudover har Lut relativt lav cytotoksicitet, n\u00e5r det g\u00e6lder om at h\u00e6mme spredningen af forskellige tumorceller og fremkalde deres apoptose, hvilket g\u00f8r det til et potentielt antitumorl\u00e6gemiddel. Det tidligere eksperiment med dette emne bekr\u00e6ftede, at Lut kan p\u00e5virke aktiviteten af PI3K\/Akt ved at opregulere ekspressionen af PETN og derefter h\u00e6mme spredningen af brystkr\u00e6ftceller. Men i betragtning af at forekomsten og udviklingen af brystkr\u00e6ft har en r\u00e6kke reguleringsmekanismer, er det stadig uklart, om Lut har andre mekanismer til at h\u00e6mme spredningen af brystkr\u00e6ftceller. Ribosomalt protein (RP) er en af hovedkomponenterne i ribosomer, der er involveret i intracellul\u00e6r proteinbiosyntese, transkription, translation og celleproliferation i andre regulatoriske molekyl\u00e6re relaterede regioner. Ribosomalt protein S12 (RPS12) er st\u00e6rkt udtrykt hos patienter med livmoderhalskr\u00e6ft, mens dets udtryk er relativt lavt i normalt v\u00e6v. Derudover er RPS12 ogs\u00e5 overudtrykt i gastrisk kr\u00e6ftv\u00e6v og kan fremme metastase af tumorceller. RPS12-genets regulerende rolle i spredningen af brystkr\u00e6ftceller er dog ikke klart defineret. C-Myc er en regulerende faktor for ribosomsyntese. Gennem cDNA-mikroarray-analyse af fibroblaster blev det konstateret, at ekspressionsniveauet af RPS12 i celler kan v\u00e6re relateret til c-Myc. Hoved\u00e5rsagen er, at tumorudviklingen og mekanismen for tidlig metastase ikke er klar. Lut, en naturlig flavonoidforbindelse, har en h\u00e6mmende effekt p\u00e5 forekomsten og udviklingen af forskellige ondartede tumorer. Selvom vores forskningsgruppe tidligere har bekr\u00e6ftet Luts h\u00e6mmende effekt p\u00e5 spredning af brystkr\u00e6ftceller, er dens specifikke mekanisme endnu ikke blevet afklaret. I denne unders\u00f8gelse fandt vi, at Lut kan nedregulere udtrykket af c-Myc ved at h\u00e6mme PI3K\/Akt\/mTOR-signalvejen og h\u00e6mme spredningen og invasionen af brystkr\u00e6ftcellerne MDA-MB-231 ved at regulere udtrykket af RPS12 negativt. Dette bekr\u00e6ftede yderligere udsigten til Lut i behandlingen af brystkr\u00e6ft.<br \/>\nRibosomer er de vigtigste organeller til proteinsyntese i celler, og eukaryote ribosomer indeholder ca. 80 RP'er. RP'er er unormalt opreguleret i forskellige tumorceller som livmoderhalskr\u00e6ft og mavekr\u00e6ft og er forbundet med spredning og invasion af tumorceller. Forskning har vist, at ekspressionsniveauet af RPS12 i mavekr\u00e6ftceller er betydeligt h\u00f8jere end i normale celler, hvilket tyder p\u00e5, at RPS12 kan v\u00e6re involveret i forekomsten og udviklingen af mavekr\u00e6ft. Denne unders\u00f8gelse viste, at RPS12 ogs\u00e5 var st\u00e6rkt udtrykt i brystkr\u00e6ftcellelinjer, hvilket var i overensstemmelse med andre tumorer. Men n\u00e5r brystkr\u00e6ftceller blev behandlet med Lut, blev ekspressionsniveauet af RPS12 reduceret betydeligt. Det blev bekr\u00e6ftet, at RPS12 ogs\u00e5 deltog i reguleringen af Lut p\u00e5 brystkr\u00e6ftceller. For yderligere at afklare RPS12's rolle i migration og invasion af brystkr\u00e6ftceller testede vi ogs\u00e5 effekten af at d\u00e6mpe RPS12 p\u00e5 celleinvasion og -migration. Resultaterne viste, at d\u00e6mpning af RPS12 kunne h\u00e6mme invasionen og migrationen af brystkr\u00e6ftceller betydeligt. Dette indikerer ogs\u00e5, at det h\u00f8je udtryk af RPS12 i brystkr\u00e6ft kan bidrage til tumorudvikling og metastase. c-Myc-genet er i \u00f8jeblikket kendt som et proto-onkogen, der effektivt kan fremme celledeling og -spredning og er t\u00e6t forbundet med dannelsen og udviklingen af forskellige ondartede tumorer. Forskning har vist, at MicroRNA-184 i tyktarmskr\u00e6ft kan h\u00e6mme spredningen af tyktarmskr\u00e6ftceller og fremme celleapoptose ved at nedregulere ekspressionsniveauet for c-Myc. Derudover har c-Myc ogs\u00e5 en lignende fremmende effekt p\u00e5 celleproliferation i mavekr\u00e6ftceller. Denne unders\u00f8gelse viste, at c-Myc ogs\u00e5 er st\u00e6rkt udtrykt i brystkr\u00e6ftcellelinjer, og nedregulering af c-Myc-ekspression kan yderligere h\u00e6mme ekspressionen og transkriptionen af RPS12 i celler, hvilket yderligere kan h\u00e6mme spredningen af brystkr\u00e6ftceller. I betragtning af c-Myc's rolle i andre tumorceller er det endnu en gang blevet p\u00e5vist, at Lut kan ud\u00f8ve sin virkning ved at p\u00e5virke udtrykket af c-Myc\/RPS12.<br \/>\nPI3K\/Akt er en enzymaktiv intracellul\u00e6r signalvej, der kan deltage i processer som tumorv\u00e6kst og -differentiering. Unders\u00f8gelser har vist, at PI3K\/Akt-signalvejen ofte er unormalt aktiveret under spredning og migration af brystkr\u00e6ftceller. I denne unders\u00f8gelse blev b\u00e5de Akt-h\u00e6mmeren LY294002 og mTOR-h\u00e6mmeren rapamycin brugt til at h\u00e6mme fosforyleringsniveauerne af mTOR og S6K betydeligt. Rapamycin havde dog ingen signifikant effekt p\u00e5 Akt-phosphoryleringsniveauerne, da mTOR er et downstream-molekyle af PI3K\/Akt og derfor ikke har nogen effekt p\u00e5 Akt-phosphorylering. Desuden kan PI3K\/Ak- og mTOR-h\u00e6mmere reducere udtrykket af RPS12 og c-Myc i celler betydeligt og dermed h\u00e6mme celleproliferationen. Disse resultater tyder p\u00e5, at RPS12 og c-Myc ogs\u00e5 kan v\u00e6re downstream-molekyler i PI3K\/Akt\/mTOR-signalvejen og deltage i reguleringen af Lut p\u00e5 spredning og migration af brystkr\u00e6ftceller.<br \/>\nSammenfattende kan Lut h\u00e6mme spredningen og invasionen af brystkr\u00e6ft MDA-MB-231-celler ved at h\u00e6mme PI3K\/Akt\/mTOR-signalvejen og derved nedregulere udtrykket af c-Myc og derefter regulere RPS12 negativt. Disse resultater tyder p\u00e5, at RPS12 og c-Myc kan v\u00e6re potentielle m\u00e5l for behandling af brystkr\u00e6ft, og Lut kan spille en antitumorrolle ved at regulere udtrykket af RPS12 og c-Myc.
