Li et al. isolerede og identificerede monotrichothecener fra S. chartarum WGC-25C-6 isoleret fra svampe. Blandt dem udviser forbindelser som mytoxin A og satrotoxin G betydelig antitumoraktivitet mod HCT-116, HepG2, BGC-823 og andre cellelinjer med IC50&lt;0,01 \u03bc mol\/L. Yang et al. identificerede seks diterpenoidforbindelser fra den marine afledte papirdruesvamp J403-SS6. CPCC 401591 til dybdeg\u00e5ende forskning. Ved at kombinere OSMAC-teknologi, LC-MS\/MS-molekyl\u00e6r netv\u00e6rksanalyse og PSM-forbindelsens UV-absorptionsegenskaber blev ethylacetatfaseseparation og oprensning af faste risfermenteringsprodukter fra stamme CPCC 401591 unders\u00f8gt. Samtidig sigter vi mod at udtr\u00e6kke og isolere antitumoraktive molekyler fra denne stamme kombineret med tumorcellescreeningsmodeller s\u00e5som den humane levercancercellelinje HepG2. Det g\u00f8r OSMAC-teknologien til en billig, effektiv, alsidig og relativt enkel forskningsmetode til aktivering af mikrobiel sekund\u00e6r metabolisme. Denne unders\u00f8gelse fokuserer p\u00e5 stammen CPCC 401591 og bruger OSMAC-strategien til at unders\u00f8ge virkningerne af PDB-, BY-, F2- og risfermenteringsmedier p\u00e5 svampens sekund\u00e6re metabolitter. Baseret p\u00e5 m\u00e6ngden og udbyttet af sekund\u00e6re metabolitter i stammen blev ris-kulturmediet bestemt til at v\u00e6re det optimale fermenteringsmedium, hvilket indikerer, at OSMAC-teknologien forbedrer effektiviteten af udvinding af sekund\u00e6re metabolitter i denne stamme betydeligt. Samtidig anvendte denne unders\u00f8gelse GNPS-molekyl\u00e6r netv\u00e6rksanalysestrategi til at analysere ethylacetatfasen af fermenteringsekstraktet af stamme CPCC 401591. Analytens sekund\u00e6re massespektrometri-data blev sammenlignet med de sammensatte massespektrometri-data i databasen for hurtigt at identificere de sammensatte kategorier, der er indeholdt i analytpr\u00f8ven, og yderligere hj\u00e6lpe med den effektive opdagelse af bioaktive molekyler.<\/p>\n<p>PSM-forbindelser er i \u00f8jeblikket kun isoleret fra svampe af sl\u00e6gterne Botrytis og Umeda, hvilket g\u00f8r dem til karakteristiske molekyler for disse to typer svampe. Forbindelsen stackybotrin D udviser betydelig anti-HIV-aktivitet med EC50-v\u00e6rdier fra 6,2 til 23,8 \u03bcM mod HIV-1 wt, HIV-1 RT-K103N og andre mutantstammer. Jia et al. fandt, at IC50-v\u00e6rdierne for bistachybotrysin K p\u00e5 tumorcellelinjer som HCT116 og NCI-H460 varierede fra 1,1 til 4,7 \u03bc M. Dette studie unders\u00f8gte de sekund\u00e6re metabolitter i stammen Stachybotrys sp. CPCC 401591. Derfor kan der findes flere PSM-molekyler ved yderligere at \u00f8ge fermenteringsudbyttet, optimere fermenteringsbetingelserne og kombinere HPLC-UV-MS-teknologi. P\u00e5 nuv\u00e6rende tidspunkt har vi afsluttet helgenomsekventeringen af stammen Stachybotrys sp. CPCC 401591 og lokaliseret to mulige biosyntetiske genklynger psd1 og psd2 af PSM-forbindelser ved hj\u00e6lp af genome mining-metoder. Begge genklynger indeholder gener, der koder for polyketidsyntase og sesquiterpensyntase, mens denne genklynge koder for rigelige postmodificerende enzymer (herunder oxidase P450s, kortk\u00e6det dehydrogenase og acetyltransferase osv.) Dette danner grundlag for vores fremtidige forskning i brugen af gen-knockout-strategier til at belyse biosyntesevejene for PSM-forbindelser og identificere de vigtigste biologiske enzymers funktioner.
