De kemiske komponenter i verbena er hovedsageligt iridoidglykosider, flavonoider, steroler, triterpener, polyfenoler, flygtige olier, tanniner, betakaroten, koffeinsyre, sukkerarter osv. De vigtigste aktive ingredienser er iridoidglykosider. Den har en k\u00f8lig natur, en bitter smag og h\u00f8rer til lever- og miltmeridianerne. Den fremmer blodcirkulationen, opl\u00f8ser blodstase, afgifter, fremmer diurese, reducerer gulsot og modvirker malaria. Klinisk bruges den til ophobning af symptomer som dysmenor\u00e9, amenor\u00e9, faryngitis, absces, \u00f8demer, gulsot og malaria. Der er mange ressourcer i verbena, bl.a. iridoidglykosider, som har forskellige virkninger som antioxidant, hepatobeskyttende, neurobeskyttende, hjerte- og hjerneisk\u00e6mibeskyttende, antiinflammatorisk osv. Kinesiske urteafkog (standardafkog) er de mest anvendte traditionelle kinesiske medicinpr\u00e6parater i klinisk praksis og ogs\u00e5 de l\u00e6ngste og mest anvendte pr\u00e6parater i den kinesiske medicins historie. Uanset om det drejer sig om afkog af traditionel kinesisk medicin, afkog af brudstykker, formelgranulat af traditionel kinesisk medicin og andre afkog af ny kinesisk medicin, tages de alle ved afkog eller vandskylning. Derfor er det af stor betydning at bruge forskning i standardafkog som den foretrukne kontrol til unders\u00f8gelse af andre kinesiske medicinpr\u00e6parater. Denne unders\u00f8gelse belyser den kvantitative overf\u00f8rselslov fra afkogsstykker til standardafkog og l\u00e6gger grunden til forskning og kvalitetsevaluering af traditionelle kinesiske medicinformuleringer af verbena.<br \/>\nDenne unders\u00f8gelse fokuserer p\u00e5 fremstilling af verbascosid, 5-hydroxyverbascosid og verbascosid i verbascosidskiver og etablerer dets UPLC-karakteristiske kort. I de indledende eksperimenter sammenlignede denne unders\u00f8gelse mobile faser som methanolvand, acetonitrilvand, acetonitril-0,1%-phosphorsyreopl\u00f8sning og acetonitril-0,05%-phosphorsyreopl\u00f8sning. Endelig blev acetonitril-0,05% fosforsyreopl\u00f8sning valgt som den mobile fase. Under de ovenn\u00e6vnte kromatografiske forhold var den kromatografiske topseparation, det teoretiske pladenummer og topformsparametrene alle gode. Denne unders\u00f8gelse anvendte et eksperimentelt system med en enkelt faktor til at unders\u00f8ge virkningerne af ekstraktionsmiddel, ekstraktionsmetode og -tid samt forholdet mellem fast stof og v\u00e6ske p\u00e5 ekstraktionseffektiviteten. Resultaterne viste, at ekstraktionseffektiviteten var h\u00f8jest, n\u00e5r ekstraktionsmidlet var 80% methanol, forholdet mellem faststof og v\u00e6ske var 1:50 (g\/mL), og tilbagesvalingsekstraktion blev udf\u00f8rt i 120 minutter. Derfor blev ovenst\u00e5ende metode brugt som ekstraktionsmetode til verbenaskiver.<br \/>\nDen overordnede kvalitetskontrol af multi-indeks-komponenter i traditionel kinesisk medicin er blevet en uundg\u00e5elig tendens i udviklingen af kvalitetskontrol af traditionel kinesisk medicin. Det karakteristiske\/fingeraftryksspektrum er en effektiv kvalitetsevalueringsmetode, og ved at kombinere det med analysemetoden med \u00e9n test og flere evalueringer kan man opn\u00e5 integritet og n\u00f8jagtighed i kvalitetsevalueringen. Denne unders\u00f8gelse etablerede et UPLC-karakteristisk kort over verbascosid-afkogsstykker, identificerede 5 almindelige toppe og identificerede 3 komponenter. Analysetiden blev v\u00e6sentligt forkortet sammenlignet med HPLC, og de kromatografiske forhold kunne bruges til bestemmelse af 5-hydroxyverbascosid, verbascosid og verbascosidindhold. Hovedkomponentanalysen blev udf\u00f8rt p\u00e5 topomr\u00e5det for karakteristiske toppe, og den omfattende evalueringsmodel for hovedkomponenter forbedrede kvalitetsevalueringen og analysemetoden for verbena yderligere.<\/p>\n<p>Indholdsbestemmelsesindekset for verbena i den kinesiske farmakop\u00e9 fra 2020 er den samlede m\u00e6ngde oleanolsyre og ursolsyre, som begge er triterpenoider med lav polaritet og lav vandopl\u00f8selighed. Forel\u00f8bige eksperimentelle forskningsresultater viser, at det samlede indhold af oleanolsyre og ursolsyre i standardafkoget af verbena er mindre end 2,0 mg\/g, og overf\u00f8rselshastigheden er mindre end 6%. Baseret p\u00e5 funktionskortet blev der derfor etableret en multievalueringsmetode til bestemmelse af indholdet af 5-hydroxyverbascosid, verbascosid og verbascosid. De pol\u00e6re komponenter i verbena er hovedsageligt cyclohexenetherterpener, blandt hvilke verbascosid og 5-hydroxyverbascosid er de vigtigste komponenter med det h\u00f8jeste indhold, og standardafkoget har en h\u00f8jere overf\u00f8rselshastighed. Overf\u00f8rselshastigheden for nogle partier overstiger 100%, mens det samlede overf\u00f8rselsomr\u00e5de for 5-hydroxyverbascosid og verbascosid ligger mellem 56,6% og 91,2%. Den mulige \u00e5rsag er, at lignende komponenter gennemg\u00e5r strukturel transformation under opvarmning, kogning og andre processer. Der er forskelle i indholdet og overf\u00f8rselshastigheden af verbascosid, 5-hydroxyverbascosid og verbascosid blandt de 18 partier af standardafkog af verbascosid, hvilket kan v\u00e6re relateret til indholdet og teksturen af afkogstykkerne. Nogle afkogningsstykker har lysere farver, l\u00e6ngere opbevaringstid og mere snavs under behandlingen, hvilket kan v\u00e6re \u00e5rsagen til de store udsving i deres indhold og overf\u00f8rselshastighed. De tre udvalgte ingredienser har et h\u00f8jt indhold af verbena og har gode farmakologiske aktiviteter inden for antioxidanter, hepatobeskyttelse, neurobeskyttelse, beskyttelse mod hjerte- og hjerneisk\u00e6mi, antiinflammatoriske og andre aspekter. De er et af de vigtigste materielle grundlag for, at verbena kan ud\u00f8ve sine farmakologiske virkninger.<\/p>\n<p>Resultaterne af den multiple evalueringsmetode med \u00e9n test viste ingen signifikant forskel i forhold til den eksterne standardmetode og kan bruges til kvantitativ analyse af flere komponenter i verbena.
