Influenzavirus tilh\u00f8rer familien Orthomyxoviridae og er opdelt i fire typer: type A, type B, type C og type D. Forebyggelse og behandling af influenza, vacciner og antivirale l\u00e6gemidler er stadig de mest effektive valg. Med hensyn til at reducere forekomsten og d\u00f8deligheden anses vacciner for at v\u00e6re den mest ideelle m\u00e5de at kontrollere influenzapandemien p\u00e5. Influenzavacciner er dog kun effektive, n\u00e5r vaccinestammen matcher den epidemiske virusstamme. De mest almindelige kliniske l\u00e6gemidler til behandling af influenza er NA-h\u00e6mmere, herunder oseltamivir, zanamivir og paramivir, samt M2-protonkanalblokkere, herunder amantadin og rimantadin. Desv\u00e6rre er nogle af de nye stammer af influenza A-virus naturligt b\u00e6rere af mutationsgener, som f\u00f8rer til resistens over for disse l\u00e6gemidler. Derfor haster det med at finde nye l\u00e6gemidler mod influenza. Selvom phenylpropanoidforbindelser ikke er hovedkomponenterne i edderkoppeduft og r\u00f8d p\u00e6on, spiller de sandsynligvis en vigtig rolle i disse to urters anvendelse mod influenza. Denne unders\u00f8gelse viste, at FDE har betydelige virkninger mod influenzavirus. H\u00f8je koncentrationer af FDE kan p\u00e5virke agglutineringen af r\u00f8de blodlegemer, hvilket tyder p\u00e5, at det kan forstyrre bindingen af influenzavirus til receptorceller, men dets virkning er ikke signifikant ved lave koncentrationer. Influenzavirus, der kommer ind i v\u00e6rtscellen, gennemg\u00e5r transkription og replikation gennem forskellige komponenter i v\u00e6rtscellen. Gennem timing-eksperimentet med l\u00e6gemiddeltils\u00e6tning fandt vi ud af, at FDE kan reducere genereringen af viralt NP-protein, hvilket sandsynligvis vil forstyrre virusreplikationsprocessen. Molekyl\u00e6re dockingresultater viste, at FDE faktisk kan binde sig til det aktive sted i NP-proteinet, og NP kan v\u00e6re et effektivt m\u00e5l for FDE. Derudover opdagede vi effekten af FDE p\u00e5 NA-aktivitet gennem neuraminidaseaktivitetsinhiberingseksperimenter, og molekyl\u00e6re dockingeksperimenter underst\u00f8tter ogs\u00e5 dette resultat, hvilket tyder p\u00e5, at FDE ogs\u00e5 kan h\u00e6mme den ekstracellul\u00e6re proces med influenzavirus. Desuden er influenzavirus i sig selv ikke den vigtigste \u00e5rsag til alvorlig sygdom og d\u00f8d. Lungev\u00e6vsskade eller mere alvorligt akut luftvejssyndrom for\u00e5rsaget af virussen er hoved\u00e5rsagen til den h\u00f8je patogenicitet og d\u00f8d af influenzavira, is\u00e6r h\u00f8jpatogene influenzavira. Vi fandt, at FDE kan reducere udtrykket af iN-OS og COX-2 i makrofager inficeret med influenzavirus og ogs\u00e5 h\u00e6mme udtrykket af IP-10 i makrofager for\u00e5rsaget af virussen, hvilket tyder p\u00e5, at FDE ogs\u00e5 kan p\u00e5virke rekrutteringen og aggregeringen af yderligere inflammatoriske celler og reducere det inflammatoriske respons. Ovenst\u00e5ende resultater indikerer, at FDE kan have mere end \u00e9t m\u00e5l for sin aktivitet mod influenzavirus. Tilstedev\u00e6relsen af flere m\u00e5l i sm\u00e5 molekyleforbindelser kan g\u00f8re dem mere effektive og mindre tilb\u00f8jelige til l\u00e6gemiddelresistens, men det g\u00f8r ogs\u00e5 ofte deres virkninger in vivo mere komplekse og kan endda have flere toksiske bivirkninger.<\/p>\n<p>Sammenfattende viste denne unders\u00f8gelse, at phenylpropanoiden FDE i edderkoppeduft og r\u00f8d p\u00e6on har anti-influenzavirusaktivitet in vitro. Ved at unders\u00f8ge FDE's indvirkning p\u00e5 influenzavirus' livscyklus blev det konstateret, at dets anti-influenzamekanisme hovedsageligt forstyrrer replikations- og frigivelsesprocessen for influenzavirus, og influenzavirus NP- og NA-proteiner er sandsynlige m\u00e5l for dets virkning.
