Under afkogningsprocessen flyder en stor m\u00e6ngde komponenter over, hvilket resulterer i flere urenheder i standardafkogningen, hvilket p\u00e5virker den kromatografiske adskillelse og analyse af effektive ingredienser. N\u00e5r man bruger opl\u00f8sningsmidler med h\u00f8j polaritet som methanol og ethanol til at behandle standardafkoget, er baseline for fingeraftryksspektret ustabilt, den kromatografiske topseparation er d\u00e5rlig, de kromatografiske toppe er uordnede, og der er mange sm\u00e5 toppe. Derfor blev separations-, oprensnings- og berigelsesmetoden for ethylacetatekstraktion unders\u00f8gt i forbehandlingsmetoden for standardafkogstestpr\u00f8ver for at reducere interferensen af urenheder og sammenlignet med konventionelle behandlingsmetoder. Resultaterne viste, at brug af ethylacetat som ekstraktionsmiddel kunne forbedre p\u00e5virkningen af urenhedstoppe betydeligt, sikre renhed og adskillelse af de vigtigste kromatografiske toppe og l\u00f8se problemerne med ustabil kromatografisk baseline og lille absorption af nogle kromatografiske toppe. I unders\u00f8gelsen af kromatografiske forhold blev forskellige mobilfase-elueringssystemer unders\u00f8gt, s\u00e5som acetonitril-0,1% fosforsyreopl\u00f8sning, acetonitril-0,1% eddikesyreopl\u00f8sning, methanol-0,1% myresyreopl\u00f8sning, methanol-0,1% eddikesyreopl\u00f8sning osv. Resultaterne viste, at brug af methanol-0,1%-eddikesyreopl\u00f8sning som elueringssystem resulterede i et h\u00f8jere antal kromatografiske toppe og bedre samlet adskillelse. I betragtning af etableringen af ultrah\u00f8jtydende v\u00e6skekromatografiske fingeraftryksspektre er valget af kromatografiske kolonner afg\u00f8rende. Derfor unders\u00f8gte dette eksperiment kromatografiske kolonner med forskellige partikelst\u00f8rrelser og m\u00e6rker og valgte i sidste ende Agilent ZORBAX SB C18-kromatografikolonnen (2,1 mm \u00d7 150 mm, 1,8 \u03bc m) med den bedste topform og opl\u00f8sning af hver delt fingeraftrykstop.<br \/>\nForskning har vist, at bladene fra Bu Zha indeholder mange flavonoider. Xiao et al. identificerede 31 forbindelser fra Bu Zha Ye ved hj\u00e6lp af UPLC-Q-TOF-MS\/MS-teknologi, herunder 28 flavonoider og 3 fenoliske syrer. Farmakologiske unders\u00f8gelser har vist, at flavonoider er et vigtigt grundlag for Bu Zha Ye's ford\u00f8jelses-, varme- og fugtlindrende virkninger. Repr\u00e6sentative komponenter som vitexin, isovitexin og narcissin har et h\u00f8jt indhold i Bu Zha Ye og en vis opl\u00f8selighed i vand. Derfor bruger Bu Zha Ye-standardafkog vitexin, isovitexin og narcissin som indikatorer for indholdsbestemmelse og etablerer en metode til indholdsbestemmelse for Bu Zha Ye-afkog baseret p\u00e5 afkoget for at vejlede unders\u00f8gelsen af overf\u00f8rslen af standardafkogv\u00e6rdier.<br \/>\nBu Zha Ye-medicinske urter er hovedsageligt distribueret i Guangdong, Guangxi og Hainan og h\u00f8rer til de autentiske medicinske urter, der produceres i Guangdong. Der blev indsamlet 15 partier Bu Zha Ye-medicinske urter fra de tre f\u00f8rn\u00e6vnte produktionsomr\u00e5der, hvilket viser tilstr\u00e6kkelig repr\u00e6sentativitet. Ud fra bestemmelsesresultaterne af 15 partier af standardafkogsekstrakt, ekstrakt og 15 partier af Bu Zha Ye-afkogsstykker for tre flavonoider kan det ses, at der er visse forskelle mellem pr\u00f8verne fra forskellige partier, hvor maksimums- og minimumsv\u00e6rdierne adskiller sig ca. to gange. Denne forskel er relateret til kvaliteten af selve de medicinske materialer og opfylder kravene i de tekniske krav. Der er ingen \u00e5benlyse unormale v\u00e6rdier.<br \/>\nUd fra resultaterne af bestemmelsen af overf\u00f8rselshastigheden varierede den gennemsnitlige overf\u00f8rselshastighed for de tre flavonoider fra 40% til 60%, hvor vitexin havde den h\u00f8jeste overf\u00f8rselshastighed, efterfulgt af narcissin og isovitexin, og den samlede overf\u00f8rselshastighed var relativt stabil. Forskningsresultaterne for fingeraftryksspektret viste, at der var en vis \u00e6ndring i andelen af toppe fra Bu Zha Ye-afkogsstykker til standardafkog. OPLS-DA-analysen identificerede fem signifikant forskellige kromatografiske toppe, nemlig top 1, 2, 3, 4 og 6. Bortset fra top 3, som ikke blev identificeret, var alle andre kromatografiske toppe kendte. Blandt dem var top 1, 2 og 4 fenoliske syrekomponenter. Derfor formodes peak 3 at v\u00e6re en fenolsyre. Fenoliske syrekomponenter er en s\u00e6rlig tilstedev\u00e6relse i planter, som kan tjene som forstadier til syntese af andre forbindelser og ogs\u00e5 kan hydrolyseres fra forskellige forbindelser. Derfor anses den betydelige stigning i det relative topomr\u00e5de for fenoliske syrekomponenter i standardafkoget af Bu Zha Ye for at skyldes hydrolyse af andre komponenter under afkogningsprocessen. Strukturen af top 6 isovitexin ligner strukturen af top S isovitexin, med lille forskel i polaritet, men der er en betydelig forskel i overf\u00f8rselshastighed.
