Diosgenin er en slags naturligt steroidsaponin fra Dioscorea-planter, som har god antitumoraktivitet. Moderne farmakologi har fundet ud af, at Solanum nigrum har forskellige biologiske aktiviteter, hvoraf de fleste er koncentreret inden for antitumor. Unders\u00f8gelser har vist, at dets aktive ingrediens, diosgenin, kan ud\u00f8ve antitumoreffekter ved at regulere forskellige cellul\u00e6re signalveje og er t\u00e6t forbundet med mange vigtige molekyl\u00e6re m\u00e5l som cellev\u00e6kst, differentiering og apoptose.<br \/>\nGennem analyse af TCMSP-databasen for netv\u00e6rksfarmakologi screenede denne unders\u00f8gelse syv effektive aktive ingredienser, nemlig fraxinol, \u03b2-caroten, sitosterol, diosgenin, solanin, kolesterol og quercetin. Krydset blev opn\u00e5et ved at kortl\u00e6gge de relevante m\u00e5l for de aktive ingredienser i Solanum nigrum med introduktionslinjesoftwaren VENNY 2.1 for brystkr\u00e6ftsygdomme. Samtidig viste PPI-netv\u00e6rksanalysen, at der var et t\u00e6t forhold mellem disse krydsningsm\u00e5l. Unders\u00f8gelser har vist, at quercetin, australsk solanin og sitosterol har farmakologiske virkninger, s\u00e5som at h\u00e6mme tumorcelleproliferation og inducere tumorcelleapoptose. Diosgenin kan fremme apoptose af brystkr\u00e6ft MCF-7-celler og Skp2-celler. Ud fra dette kan man se, at de aktive ingredienser i Solanum nigrum, s\u00e5som quercetin, solanin, sitosterol og dioscin, kan ud\u00f8ve antitumoreffekter.<br \/>\nIf\u00f8lge netv\u00e6rksdiagrammet for sammensatte m\u00e5l blev der opn\u00e5et 110 m\u00e5l efter sk\u00e6ringspunktet mellem Solanum nigrum og sygdomsm\u00e5l. Yderligere screening viste, at ERBB2, EGFR, KIT, SRC, ESR1, AKT1, MAPK1, PIK3CA osv. var m\u00e5lproteinerne for de effektive ingredienser i Solanum nigrum til behandling af brystkr\u00e6ft. Blandt dem virker resveratrol, diosgenin og quercetin alle p\u00e5 AKT1- og EGFR-m\u00e5l, og AKT (serin\/threoninkinase) og EGFR (epidermal v\u00e6kstfaktorreceptor) spiller vigtige roller i celleproliferation, migration og metabolisme. Derfor er m\u00e5lretning mod AKT og EGFR en meget attraktiv behandlingsstrategi for brystkr\u00e6ft. Resultaterne af denne unders\u00f8gelse indikerer, at AKT1 og EGFR og andre m\u00e5l spiller en vigtig rolle i udviklingen af brystkr\u00e6ft og kan v\u00e6re potentielle m\u00e5l for Solanum nigrum i behandlingen af brystkr\u00e6ft. GO-berigelsesanalyse viser, at m\u00e5lgenerne for Solanum nigrum mod brystkr\u00e6ft involverer biologiske processer s\u00e5som positiv regulering af kinaseaktivitet, bindende protein serin \/ threonin kinaseaktivitet, positiv regulering af MAPK-kaskade, positiv regulering af PI3K-signal, ATP-binding osv. Disse n\u00f8gleprocesser spiller en vigtig rolle i Solanum nigrums behandling af brystkr\u00e6ft. KEGG-resultaterne viste, at i alt 137 signalveje blev beriget, herunder kr\u00e6ftvej, PI3K\/Akt-signalvej, EGFR-tyrosinkinaseinhibitorresistens-signalvej, ErbB-signalvej, MAPK-signalvej, Ras-signalvej, Rap1-signalvej osv.<br \/>\nMolekyl\u00e6r docking-teknologi blev brugt til at unders\u00f8ge affiniteten mellem de vigtigste aktive komponenter og m\u00e5lproteinerne i Solanum nigrum. Resultaterne viste, at de aktive komponenter som quercetin, sitosterol og diosgenin havde god bindingsaktivitet med m\u00e5let AKT1, EGFR, ESR1, SRC og MAPK, hvilket afspejler, at hovedmekanismen for de vigtigste aktive komponenter i Solanum nigrum, sitosterol og quercetin i behandlingen af brystkr\u00e6ft kan v\u00e6re t\u00e6t forbundet med de ovenn\u00e6vnte aktive komponenter og m\u00e5l. Solanum nigrum kan have en vis terapeutisk effekt p\u00e5 brystkr\u00e6ft gennem flere m\u00e5l og flere signalveje.<br \/>\nBaseret p\u00e5 resultaterne af molekyl\u00e6r dockingforskning blev den vigtigste aktive ingrediens i Solanum nigrum, Diosgenin, udvalgt til antitumoreksperimenter, og Diosgenins regulerende effekt p\u00e5 de forudsagte m\u00e5l EGFR og AKT1 blev verificeret.
