Den har farmakologiske virkninger som antiinflammatorisk, antioxidant og lipids\u00e6nkende. If\u00f8lge traditionel kinesisk medicin har Morinda officinalis en sur og s\u00f8d smag, en afbalanceret medicinsk egenskab og har den virkning, at den n\u00e6rer nyreessensen, afbalancerer yin og yang og forsinker aldring. Det kan effektivt forebygge kardiovaskul\u00e6re og cerebrovaskul\u00e6re sygdomme. Den specifikke mekanisme bag Hai Ba Ji i behandlingen af AS er dog stadig uklar. Derfor har jagten p\u00e5 nye, sikrere og mindre giftige l\u00e6gemidler til behandling af AS en positiv betydning. Som et nyt l\u00e6gemiddel og spiseligt produkt har Morinda officinalis en bred vifte af aktive ingredienser og farmakologiske virkninger, hvilket er i tr\u00e5d med de typiske egenskaber ved traditionel kinesisk medicins multi-m\u00e5l- og multi-vej-behandling af sygdomme. Denne unders\u00f8gelse brugte netv\u00e6rksfarmakologi til at forudsige de effektive ingredienser og m\u00e5l for Morinda officinalis i behandlingen af AS og konstruerede en THP-1-afledt makrofagmodel for at verificere, at Morinda officinalis fremmer intracellul\u00e6r kolesteroludstr\u00f8mning gennem PPAR \u03b3-signalvejen.<\/p>\n<p>Ved at screene forskellige databaser fandt man ud af, at 59 forbindelser, herunder flavonoider, phenylpropanoider og fenolsyrer, hovedsageligt spiller en terapeutisk rolle i behandlingen af AS. I de senere \u00e5r har flere forskningsrapporter vist, at flavonoider spiller en n\u00f8glerolle i behandlingen af SA og er de vigtigste aktive steder i plantemedicin. Sahib et al. fandt, at indholdet af catechiner, quercetin og kaempferol i frugten af Morinda officinalis var henholdsvis 53,68 mg\/g, 7,4 mg\/g og 6,4 mg\/g, hvilket rangerer blandt de tre mest kendte aktive flavonoider. Disse tre komponenter spiller en vigtig rolle i reguleringen af abnormiteter i blodlipiderne og forsinker \u00e5reforkalkning. Forskning har vist, at hovedkomponenten i Yixin Tongmai-granulat, catechiner, direkte kan aktivere PPAR \u03b3-vejen og forbedre AS gennem molekyl\u00e6r docking og in vitro- og in vivo-eksperimenter. Quercetin er almindeligt forekommende i forskellige planter med en betydelig andel i Morinda officinalis. Jia et al. og Li et al. rapporterede, at quercetin kan reducere plakindholdet i thoraxaorta i Apoe -\/- musens AS-model induceret af fedtfattig kost betydeligt. Forskning har vist, at lipidmetabolisme-relaterede signalveje, is\u00e6r PPAR-signalvejen, er t\u00e6t forbundet med lipidtransport\u00f8rer p\u00e5 cellemembranen og receptorer i kernen. Som vi alle ved, er PPAR klassiske nukleare receptorer placeret i kernen. Efter at have modtaget signalet fra second messenger \u00f8ger PPAR udtrykket af kolesteroltransport\u00f8rer p\u00e5 cellemembranen ved at aktivere nedstr\u00f8ms transkriptionsfaktorer, hvorved ophobningen af kolesterol i cellerne reduceres, og makrofager forhindres i at udvikle sig til skumceller. Derfor spiller PPAR-signalvejen en vigtig rolle i at reducere dannelsen af skumceller i lipidkerner og forsinke AS. Desuden er PPARG-genet ogs\u00e5 et vigtigt knudepunkt i kernenetv\u00e6rket i komponentm\u00e5lnetv\u00e6rket i denne unders\u00f8gelse.<\/p>\n<p>Baseret p\u00e5 ovenst\u00e5ende netv\u00e6rksfarmakologiske analyse konstruerede denne unders\u00f8gelse THP-1-afledte makrofager til at simulere skumceller i aterosklerotiske plaques in vitro. Efter at Morinda officinalis havde interveneret med THP-1-afledte makrofager, viste det sig, at det intracellul\u00e6re kolesterolindhold blev reduceret betydeligt, og at udtrykket af PPAR \u03b3 i cellerne blev opreguleret betydeligt. Inada et al. og Nerurkar et al. bekr\u00e6ftede henholdsvis den lipids\u00e6nkende effekt af Morinda officinalis gennem dyrefors\u00f8g. I en klinisk unders\u00f8gelse blev det konstateret, at Morinda officinalis kan reducere LDL-niveauerne i serum betydeligt og \u00f8ge niveauerne af lipoprotein med h\u00f8j densitet (HDL) hos storrygende personer. For nylig bekr\u00e6ftede Chong et al., at Morinda officinalis kan reducere lipidplaques i aorta betydeligt i en termisk oxideret palmeolie-di\u00e6t induceret AS-rotte-model, men den specifikke mekanisme er ikke blevet belyst. Tidligere har Lee et al. udvundet t\u00f8rret pulver af Morinda officinalis-frugt med 70% methanol og fundet ud af, at det kunne ophidse PPAR \u03b3-receptorer i C2C12-skeletmuskelceller. Det kan give n\u00e6ring til nyreessensen, afbalancere yin og yang og forebygge kardiovaskul\u00e6re og cerebrovaskul\u00e6re sygdomme. Derfor giver denne unders\u00f8gelse en ny farmakologisk mekanisme til effektiv forebyggelse og behandling af AS med Morinda officinalis.
