Men p\u00e5 grund af menneskers overforbrug af antibiotika er der opst\u00e5et bakteriel resistens. Det forventes, at det globale tab af ejendom for\u00e5rsaget af bakterielle infektioner hos mennesker i 2050 vil n\u00e5 op p\u00e5 $100 billioner. Staphylococcus aureus er en almindelig f\u00f8devareb\u00e5ren patogen bakterie, der kan for\u00e5rsage infektioner i luftvejene, ford\u00f8jelsessystemet og urinvejene. If\u00f8lge den amerikanske Food and Drug Administration (FDA) stammer over halvdelen af de kliniske l\u00e6gemidler fra naturprodukter eller deres syntetiske derivater, og ca. 200 naturlige antibiotika fra mikrobielle kilder bruges direkte som l\u00e6gemidler.<br \/>\nBaiji, ogs\u00e5 kendt som hyacintorkid\u00e9, hyacint baiji eller Ulan, har knolde, der ofte bruges som medicinske dele. Smagen er s\u00f8d, bitter og sammentr\u00e6kkende med en lidt kold karakter. Den virker sammentr\u00e6kkende, h\u00e6mostatisk, h\u00e6velsesreducerende og muskelregenererende. Almindeligvis brugt til behandling af hoste og opkastning af blod, ydre bl\u00f8dninger, s\u00e5r og toksiner, revner i huden, bl\u00f8dning fra maves\u00e5r osv. Hovedkomponenterne omfatter polysaccharider, phenanthren, phenanthrenquinon, dibenzen, triterpenoider, lignaner og organiske syrer. Farmakologiske unders\u00f8gelser har vist, at Bai Ji har forskellige biologiske aktiviteter s\u00e5som antiinflammatorisk, antifibrotisk, antitumor og immunregulering, men der er kun f\u00e5 rapporter om dens antibakterielle mekanisme. I dette eksperiment blev Staphylococcus aureus derfor taget som forskningsobjekt for at unders\u00f8ge forskellen i antibakteriel aktivitet mellem ekstraktet og den monomere forbindelse. I alt 10 forbindelser (1-10) blev isoleret og identificeret, herunder 6 phenanthrenforbindelser (1-6) og 4 coumarsyreanaloger (7-10). Opdagelsen af coumarsyre-analoger for f\u00f8rste gang i planter af Platycodon-sl\u00e6gten har vigtig vejledende betydning for den rige kemiske sammens\u00e6tning af planter i Orchidaceae-familien.<br \/>\nI udforskningen af forbindelsernes antibakterielle mekanisme blev den antibakterielle aktivitet af ekstrakter og monomere forbindelser f\u00f8rst screenet. Testresultaterne viste, at forbindelse 6 havde den bedste antibakterielle aktivitet mod Staphylococcus aureus (MIC0,01mh\/mL) blandt de testede forbindelser, og den antibakterielle virkning var dosisafh\u00e6ngig. SEM-resultaterne viste, at forbindelsen beskadigede den bakterielle cellev\u00e6g og membran, hvilket for\u00e5rsagede l\u00e6kage af intracellul\u00e6re stoffer. Desuden viste \u00e6ndringerne i AKP-enzymindholdet og fluorescensintensiteten af PI-proben yderligere, at forbindelse 6 beskadigede bakteriemembranerne. I klinisk brug kan ekstrakter af Atractylodes macrocephala fremskynde s\u00e5rheling og forhindre infektion, hvilket indikerer, at de antibakterielle egenskaber ved Atractylodes macrocephala kan v\u00e6re relateret til forbindelse 6 og andre phenanthren-derivater.<br \/>\nMed hensyn til strukturtyper er phenanthrenforbindelser naturlige produkter med helt andre strukturer end kommercielt tilg\u00e6ngelige antibiotika (\u03b2-lactamer, makrolider, quinoloner). I denne unders\u00f8gelse har phenanthren-forbindelser vist gode antibakterielle virkninger, hvilket giver inspiration til overflod af nuv\u00e6rende antibakterielle l\u00e6gemidler. Udviklingen af nye phenanthren-antibiotika forventes at l\u00f8se problemet med l\u00e6gemiddelresistente bakterier. If\u00f8lge litteraturrapporter kan selv den samme type phenanthrenforbindelser udvise forskellige antibakterielle mekanismer. For eksempel h\u00e6mmer blestricin hovedsageligt v\u00e6ksten af Staphylococcus ved at forstyrre bakteriemembranens potentiale og integritet, mens aristoloxazin C h\u00e6mmer v\u00e6ksten af Pseudomonas aeruginosa ved at inducere nedbrydning og ablation af cellev\u00e6ggen. Strukturelt set omdannes phenanthrenforbindelser let til quinoner i levende organismer, hvilket f\u00f8rer til produktion af frie semiquinonradikaler og dannelse af antibakteriel aktivitet. Det tyder p\u00e5, at det er n\u00f8dvendigt at unders\u00f8ge phenanthrenforbindelsernes antibakterielle mekanisme yderligere.
