I de senere \u00e5r er forekomsten af POF hos kvinder i den f\u00f8dedygtige alder steget, og faldet i \u00e6ggestokkens funktion har alvorligt p\u00e5virket patienternes livskvalitet og \u00f8get risikoen for hjerte-kar-sygdomme, osteoporose, metaboliske lidelser og andre relaterede sygdomme markant. Hos patienter med kr\u00e6ft og systemisk lupus erythematosus er den kliniske anvendelse af POF og cyclophosphamid (CP) t\u00e6t forbundet. CP kan forstyrre \u00e6ggestokkenes funktion gennem oxidativ stress, hormonudskillelse og inflammation. At finde en sikker og effektiv beskytter af \u00e6ggestokkenes funktion er n\u00f8glen til efterf\u00f8lgende klinisk l\u00e6gemiddeludvikling.<br \/>\nYam, som er et af de traditionelle kinesiske l\u00e6gemidler med samme oprindelse som medicin og f\u00f8devarer, indeholder rige aktive ingredienser som steroide saponiner, polysaccharider, allantoin, flavonoider og fenoliske glykosider. Mangfoldigheden i den kemiske sammens\u00e6tning giver den forskellige farmakologiske virkninger, s\u00e5som antioxidant, antiinflammatorisk, immunregulering, urinsyres\u00e6nkning, antitumor og terapeutiske virkninger p\u00e5 nyresygdomme. Forskning har vist, at udvikling og anvendelse af kinesiske l\u00e6gemidler og naturprodukter, der har samme oprindelse som f\u00f8devarer og medicin, har stor betydning for beskyttelsen af \u00e6ggestokkene. Allantoin (ALL) er den vigtigste aktive ingrediens i yams, som har funktioner som antistress, antiinflammatorisk, regulering af tarmens mikrobiota og fremme af cellev\u00e6kst. I tidligere unders\u00f8gelser fandt forskerholdet, at yams og ALL har \u00f8strogenlignende virkninger gennem ekspression af \u00f8strogenreceptor alfa og G-proteinkoblede \u00f8strogenreceptorer i livmoderv\u00e6v. Naturlige plante\u00f8strogener har en positiv effekt p\u00e5 udviklingen af follikler i \u00e6ggestokkene hos rotter. I de sidste par \u00e5rtier er beskyttelsen af forplantningsevnen og genoprettelsen af \u00e6ggestokkenes funktion blevet meget vigtig i kr\u00e6ftbehandlingen, da dette behandlingsregime kan forbedre livskvaliteten for kvindelige kr\u00e6ftpatienter. Derudover kan CP ogs\u00e5 p\u00e5virke st\u00f8rre organer som hjerte og nyrer ved at aktivere oxidative stressmark\u00f8rer og overforbrug af antioxidante forsvarsmekanismer. Derfor er et af fokuspunkterne i denne unders\u00f8gelse at forbedre CP's skader p\u00e5 organer ved at regulere oxidativ stress.<\/p>\n<p>CP's toksicitet er relateret til produktionen af en af dets metabolitter, acrolein. Efter CP-behandling udviser kr\u00e6ftpatienter et betydeligt fald i glutathion (GSH), hvilket kan v\u00e6re et resultat af bindingen af acrolein. GSH er et tripeptid og en vigtig antioxidant og opsamler af frie radikaler i kroppen. Samtidig \u00e6ndrer CP ogs\u00e5 aktiviteten af andre endogene antioxidanter i sundt ovariev\u00e6v, s\u00e5som SOD, hvilket fremmer intracellul\u00e6r skade p\u00e5 frie radikaler i ovarieceller. I denne unders\u00f8gelse beskyttede ALL rotternes \u00e6ggestokke betydeligt mod oxidativ stress for\u00e5rsaget af CP. ALL er en metabolit, der produceres ved nedbrydning af puriner, og som findes i kroppen hos dyr og planter. Det kan fremme hudcellev\u00e6kst og s\u00e5rheling, stimulere fibroblastv\u00e6kst og har \u00f8strogenlignende effekter. Derfor fokuserer denne unders\u00f8gelse, samtidig med at den udforsker dets beskyttende virkninger mod organskader, ogs\u00e5 p\u00e5 dets antioxidante virkninger og virkningsveje. I denne unders\u00f8gelse viste det sig, at 70 mg\/kg og 140 mg\/kg allantoin opretholdt GSH Px- og SOD-aktiviteten i \u00e6ggestokkene og h\u00e6mmede MDA betydeligt, hvilket beskyttede rotternes \u00e6ggestokke mod CP-induceret oxidativ stress, der for\u00e5rsagede skader p\u00e5 \u00e6ggestokkene.<\/p>\n<p>Den nukleare transkriptionsfaktor Nrf2 tilh\u00f8rer CNC leucine zipper transkriptionsaktivatorfamilien, som beskytter proteinkodende gener ved at inducere antioxidantaktivitet og regulere oxidative deoxygeneringsreaktioner i det intracellul\u00e6re milj\u00f8. Nrf2 aktiverer downstream-antioxidantproteiner og -enzymer ved at binde sig til antioxidantresponselementer (ARE) og beskytter mod oxidativ stress. I denne unders\u00f8gelse fandt vi et fald i Nrf2-ekspressionen og en stigning i oxidativ stress i POF-rotternes \u00e6ggestokke; ALL regulerer Nrf2-niveauet ved en bestemt dosis. Forskning har vist, at udtrykket af downstream-m\u00e5lgener (HO-1, NQO1 osv.) er i overensstemmelse med Nrf2-nuklear translokation, hvilket ikke kun bekr\u00e6fter inddragelsen af Kelch like as social protein 1 (Keap1) - Nrf2 ARE-vejen, men ogs\u00e5 antyder, at HO-1 og NQO1 spiller en kritisk rolle i opretholdelsen af redoxhomeostase. HO-1 er en undertype af heme oxygenase, som let stimuleres og induceres, og som spiller en indirekte antioxidantrolle. NQO1 kan fjerne superoxidanionradikaler og afhj\u00e6lpe den overdrevne ophobning af reaktive iltarter (ROS), der opreguleres af oxidativ stress. I vores unders\u00f8gelse viste POF-rotte-modellen ikke signifikant ekspression af NQO1 i \u00e6ggestokkene. Men efter kontinuerlig administration i 14 dage \u00f8gede det positive l\u00e6gemiddel EV og ALL ekspressionsniveauet af NQO1 signifikant ved en dosis p\u00e5 70 mg\/kg, hvilket tyder p\u00e5, at de ogs\u00e5 kan spille en positiv rolle i ovarieskade-stress. Dette resultat kan v\u00e6re relateret til stabiliteten, de to aktive steder og den ikke-enzymatiske aktivitet af NQO1. Det er muligt, at CP-stimulering ikke aktiverede NQO1, mens ALL kan binde sig til det aktive sted ved en bestemt dosis. Derfor er der behov for yderligere forskning og udforskning. POF-rotte-modellen reducerede signifikant udtrykket af HO-1 i \u00e6ggestokkene, mens det positive l\u00e6gemiddel EV og ALL viste en signifikant forbedring af HO-1-ekspressionen ved en dosis p\u00e5 140 mg\/kg. HO-1 er et stressresponsivt protein, der katalyserer nedbrydningen og omdannelsen af h\u00e6mproteiner til bilirubin. Produkterne fra dets nedbrydningsmetabolisme har antioxidante, antiapoptotiske og immunmodulerende virkninger. Derfor kan HO-1 spille en beskyttende rolle under stress, hvilket tyder p\u00e5, at ALL kan forbedre skader p\u00e5 \u00e6ggestokkene hos POF-rotter ved at regulere HO-1 til at ud\u00f8ve antioxidante stresseffekter.
