Sm., en ortop\u00e6disk vanddrageplante. Den er varm af natur, bitter i smagen og h\u00f8rer til lever- og nyremeridianerne. Den virker helbredende og smertelindrende, styrker nyrerne og knoglerne og fordriver vind og pletter. Moderne farmakologiske og kliniske unders\u00f8gelser har vist, at knoglefragmenttonika har virkninger som f.eks. antiosteoporose, fremmer heling af brud, beskytter nyrerne, er antiinflammatorisk og fremmer tandv\u00e6kst. Unders\u00f8gelser af den kemiske sammens\u00e6tning har vist, at de vigtigste aktive ingredienser i knoglefragmenttonika er flavonoider, triterpenoider, phenylpropanoider og phenoler. Et stort antal unders\u00f8gelser har vist, at knoglemarvstonika har betydelige terapeutiske virkninger i behandlingen af knoglerelaterede sygdomme, og in vitro-eksperimenter har vist, at knoglemarvstonika kan fremme osteoblasternes osteogene differentieringsevne. Der har dog ikke v\u00e6ret nogen rapporter om de specifikke aktive ingredienser i knoglemarvstonika. Alkalisk fosfatase (ALP) er en biomark\u00f8r for differentiering og modning af osteoblaster, og \u00e6ndringer i ALP's aktivitetsniveau kan bruges til at bestemme osteoblasternes differentieringsevne. Derfor analyserede dette eksperiment forholdet mellem UPLC-fingeraftryk og ALP-aktivitetsspektrum for humane osteosarkom MG63-celler og unders\u00f8gte hovedkomponenterne i knoglefragmenttonika, der bruges til osteoblaster.<br \/>\nEvalueringsmetoden for traditionel kinesisk medicins spektrumeffektivitet er at kombinere den kemiske komponents fingeraftryk med dens farmakologiske virkninger, s\u00e5 de kemiske komponenter i fingeraftrykket afspejler de tilsvarende farmakologiske virkninger, og afklare forholdet mellem de karakteristiske toppe i fingeraftrykket og de farmakologiske virkninger, bestemme det tilsvarende farmakologiske stofgrundlag og dermed udf\u00f8re kvalitetskontrol og farmakologisk evaluering af traditionel kinesisk medicin.<br \/>\nDenne unders\u00f8gelse etablerede UPLC-fingeraftryksspektre af r\u00e5 og varme knoglefragmenttilskud og identificerede deres kemiske komponenter ved hj\u00e6lp af UPLC-Q-TOF-MS-teknologi. Der var 12 f\u00e6lles toppe mellem forskellige partier af knoglefragmenttilskud og varme produkter, og ligheden var st\u00f8rre end 0,9, hvilket indikerer h\u00f8j lighed og uklar differentiering mellem de to. ALP er en biomark\u00f8r for osteoblastdifferentiering og -modning og er en af de biokemiske indikatorer for knogledannelse. Osteoblasternes differentieringsevne kan bestemmes ud fra \u00e6ndringer i ALP's aktivitetsniveau. Tidligere unders\u00f8gelser har vist, at tilskud med knoglefragmenter kan fremme osteoblasternes differentiering. I denne unders\u00f8gelse blev tilskud med knoglefragmenter og skoldninger anvendt p\u00e5 MG63-celler for at analysere deres effekt p\u00e5 ALP-aktiviteten. Resultaterne viser, at det samme parti knoglefragmenter i h\u00f8j grad kan fremme udskillelsen af ALP fra MG63-celler efter sandskoldning. For at unders\u00f8ge de aktive ingredienser i knoglefragmentering og -reparation, der fremmer MG63's osteogene differentiering, blev der udf\u00f8rt en spektral korrelationsanalyse mellem ALP-aktivitet og karakteristiske toppe kalibreret ved hj\u00e6lp af fingeraftryksspektre. For at foretage en omfattende analyse af det spektrale effektforhold mellem de karakteristiske toppe i knoglefragmentplaster-fingeraftryksspektret og ALP-aktiviteten blev der i dette eksperiment anvendt tre metoder: gr\u00e5 korrelationsanalyse, bivariat korrelationsanalyse og regressionsanalyse med mindste kvadraters metode. Det blev konstateret, at peak 4, peak 5 og peak 12 har en h\u00f8j korrelation med ALP-aktivitet og er positivt korrelerede, bidrager mest til ALP-aktivitet og er vigtige variabler, der p\u00e5virker ALP-aktivitet.<br \/>\nUPLC-Q-TOF\/MS-teknologi har egenskaberne st\u00e6rk selektivitet, h\u00f8j f\u00f8lsomhed, enkel pr\u00f8veforbehandling, h\u00f8j n\u00f8jagtighed og h\u00f8j opl\u00f8sning, som mere omfattende og hurtigt kan identificere de kemiske komponenter i knoglefragmenter. Denne unders\u00f8gelse brugte UPLC-Q-TOF\/MS i negativ iontilstand til at identificere 40 komponenter i tonika med knoglefragmenter. Den identificerede komponent, der bidrager v\u00e6sentligt til ALP-aktiviteten, er peak 4 caffeic acid-4-O - \u03b2 - D-glucopyranoside; Peak 5 catechins 7-glucoside; Peak 12 naringin og dens aktivitet blev valideret. Naringin er en flavonoidforbindelse, og moderne farmakologisk forskning har vist, at naringin kan bruges til at behandle eller forebygge forskellige sygdomme, s\u00e5som osteoporose, antioxidant, antitumor og forbedring af myokardieskader. Eksisterende forskning viser ogs\u00e5, at naringin er en vigtig aktiv ingrediens i tonika til knoglefragmenter. De farmakologiske aktiviteter af koffeinsyre-4-O - \u03b2 - D-glukopyranosid og catechin 7-glukosid er endnu ikke blevet rapporteret, og yderligere forskning er berettiget.
