Det er en vigtig olieafgr\u00f8de i Middelhavsomr\u00e5det, og Gansu og Yunnan er de vigtigste omr\u00e5der for olivendyrkning i Kina. Olivenblade er et billigt biprodukt ved dyrkning af oliventr\u00e6er. Hvert oliventr\u00e6 har brug for at trimme ca. 25 kg blade hvert \u00e5r, og den samlede v\u00e6gt af h\u00f8stede blade udg\u00f8r 10% af den samlede v\u00e6gt af oliven. Olivenblade indeholder forskellige bioaktive forbindelser. S\u00e5som oleanolsyre, tj\u00f8rninsyre, ursolsyre, oleuropein, hydroxytyrosol osv.<br \/>\nForskning har vist, at oleuropein findes i alle dele af oliventr\u00e6er, og at det h\u00f8jeste indhold findes i olivenblade. I t\u00f8rre olivenblade kan indholdet af oleuropein n\u00e5 op p\u00e5 10% til 17%. Som vist i figur 1 hydrolyseres oleuropein til hydroxytyrosol, glukose og olensyre. Blandt dem udviser hydroxytyrosol bedre biologisk aktivitet end oleuropein. P\u00e5 nuv\u00e6rende tidspunkt omfatter hydrolysemetoderne for oleuropein hovedsageligt syrehydrolyse, alkalisk hydrolyse og enzymatisk hydrolyse. Syrehydrolyse og alkalisk hydrolyse kr\u00e6ver st\u00e6rke syrer, st\u00e6rke baser og milj\u00f8er med h\u00f8j temperatur og strenge reaktionsbetingelser. Liu et al. brugte cellulase som f\u00f8devaretils\u00e6tningsstof til at bryde glykosidbindinger og hydrolysere oleuropein for at opn\u00e5 hydroxytyrosol. P\u00e5 grund af cellulasens termiske ustabilitet kan den ikke opretholde h\u00f8j aktivitet kontinuerligt ved enzymets optimale temperatur, hvilket resulterer i lav enzymatisk hydrolyseeffektivitet. Derfor fors\u00f8ger vi at finde et nyt gr\u00f8nt opl\u00f8sningsmiddel til at erstatte bufferopl\u00f8sningen, som kan \u00f8ge opl\u00f8seligheden af oleuropein og forbedre cellulasens stabilitet i langtidsreaktioner og dermed forbedre den enzymatiske hydrolyseeffektivitet af oleuropein.<br \/>\nDybe eutektiske opl\u00f8sningsmidler (DES) er homogene og gennemsigtige blandinger dannet ved h\u00f8jtemperaturomr\u00f8ring af hydrogenbindingsacceptorer (HBA) og hydrogenbindingsdonorer (HBD) med lavere smeltepunkter sammenlignet med de enkelte komponenter. P\u00e5 grund af sine fremragende fysiske og kemiske egenskaber, herunder lav br\u00e6ndbarhed, lav flygtighed, nem fremstilling, h\u00f8j opl\u00f8selighed og h\u00f8j kompatibilitet med enzymer, bruges DES i stigende grad som et ikke-giftigt ekstraktionsmiddel og opl\u00f8sningsv\u00e6ske. Det vigtigste er, at enzymer er tilb\u00f8jelige til at blive deaktiveret i organiske opl\u00f8sningsmidler, og DES som opl\u00f8sningsmiddel kan forhindre dette f\u00e6nomen i at opst\u00e5. Det g\u00f8r det ogs\u00e5 muligt for DES at erstatte organiske opl\u00f8sningsmidler og inkubere cellulaser. DES, der best\u00e5r af naturlig HBA og HBD, kaldes NADES, som er sikrere og gr\u00f8nnere end konventionel DES. NADES har f\u00e5et stor opm\u00e6rksomhed som et gr\u00f8nt opl\u00f8sningsmiddel inden for enzymkatalyserede reaktioner. NADES er med succes blevet anvendt i specifikke enzymatiske reaktioner og har haft en positiv effekt p\u00e5 b\u00e5de enzymaktivitet og -stabilitet. Ovenst\u00e5ende forskningsresultater viser, at NADES har potentiale til at blive et godt opl\u00f8sningsmiddel til cellulasehydrolyse af oleuropein.<br \/>\nDenne unders\u00f8gelse fremstillede seks typer NADES ved hj\u00e6lp af cholinklorid eller betain som HBA og forskellige typer polyoler som HBD og unders\u00f8gte deres virkning p\u00e5 cellulasehydrolyse af oleuropein.
