I 2020 var der 10,06 millioner nye kr\u00e6fttilf\u00e6lde hos m\u00e6nd og 5,53 millioner d\u00f8dsfald p\u00e5 verdensplan, herunder 1,41 millioner nye PCa-tilf\u00e6lde og 380000 d\u00f8dsfald, med et forhold mellem d\u00f8delighed og forekomst p\u00e5 27%. I Kina var der 120000 nye PCa-sager og 50000 d\u00f8dsfald med en PCa-d\u00f8delighed i forhold til incidens p\u00e5 41,7%. If\u00f8lge forskningsdata fra American Cancer Society i 2023 ligger forekomsten af PCa p\u00e5 f\u00f8rstepladsen blandt mandlige maligne tumorer og tegner sig for ca. 29%. P\u00e5 nuv\u00e6rende tidspunkt omfatter de vigtigste behandlingsmuligheder for PCa i klinisk praksis androgen deprivationsterapi, radikal prostatektomi, str\u00e5lebehandling, kemoterapi og molekyl\u00e6r m\u00e5lrettet terapi. Disse behandlingsmetoder har dog mange bivirkninger og er tilb\u00f8jelige til l\u00e6gemiddelresistens, hvilket resulterer i d\u00e5rlig prognose og tilbagefald eller progression hos nogle patienter. Derfor er det af stor betydning at s\u00f8ge efter sikre og effektive nye l\u00e6gemidler til behandling af PCa.<br \/>\nSophoridin (SRI) er et tetracyklisk quinolinalkaloid, der hovedsageligt er isoleret fra Sophora alopecuroides L. Forskning har vist, at SRI har mange farmakologiske virkninger, herunder antiinflammatoriske, antivirale og antitumorvirkninger. I 2005 blev injektion af sophocarpinhydrochlorid godkendt af State Food and Drug Administration til klinisk behandling af maligne trofoblastiske tumorer. P\u00e5 nuv\u00e6rende tidspunkt er SRI blevet afpr\u00f8vet til behandling af levermetastaser fra gastrointestinale tumorer. Samtidig har et stort antal rapporter bekr\u00e6ftet, at SRI har en god h\u00e6mmende effekt p\u00e5 kolorektal cancer, lungecancer, bugspytkirtelcancer og andre tumorer. SRI's indvirkning p\u00e5 PCa og dens mulige virkningsmekanisme er dog stadig uklar.<br \/>\nNetv\u00e6rksfarmakologi er en ny disciplin baseret p\u00e5 klassisk farmakologi, bioinformatik og computervidenskabelige teorier, der i vid udstr\u00e6kning anvendes til l\u00e6gemiddeludvikling og forskning i l\u00e6gemidlers virkningsmekanismer. Denne artikel unders\u00f8ger virkningerne af SRI p\u00e5 spredning og apoptose af DU-145-celler gennem in vitro-celleeksperimenter og kombinerer netv\u00e6rksfarmakologi og molekyl\u00e6re dockingteknikker for at udforske dets mulige virkningsmekanisme for at give teoretisk grundlag og datast\u00f8tte til den kliniske behandling af PCa med SRI.<\/p>\n<p><img loading=\"lazy\" alt=\"\" width=\"729\" height=\"412\" \/><\/p>\n<p>PCa er den mest almindelige ondartede tumor blandt midaldrende og \u00e6ldre m\u00e6nd, som er tilb\u00f8jelig til l\u00e6gemiddelresistens og metastase, hvilket f\u00f8rer til d\u00e5rlig prognose og h\u00f8j tilbagefaldsrate. Derfor er det blevet meget presserende at finde nye behandlingsformer til PCa. De naturlige kemiske aktive ingredienser i traditionel kinesisk medicin spiller en vigtig rolle i forebyggelse og behandling af ondartede tumorer. SRI er hovedsageligt et aktivt alkaloid isoleret fra den traditionelle kinesiske medicin Sophora alopecuroides, som har forskellige farmakologiske aktiviteter, blandt hvilke antitumoraktivitet er s\u00e6rlig fremtr\u00e6dende. I 2005 blev sophocarpine hydrochloride injection godkendt af China Food and Drug Administration som et l\u00e6gemiddel mod kr\u00e6ft. SRI har vist gode antitumoreffekter p\u00e5 mavekr\u00e6ft, leverkr\u00e6ft, lungekr\u00e6ft, tyktarmskr\u00e6ft, bugspytkirtelkr\u00e6ft og andre tumorer in vivo og in vitro. SRI kan h\u00e6mme spredningen og invasionen af forskellige tumorceller, fremme apoptose og autofagi af kr\u00e6ftceller. Dets funktion er relateret til veje som PI3K\/AKT, Wnt\/B-catenin, MAPK\/ERK og cellecyklus. Hvad ang\u00e5r den nuv\u00e6rende forskning, er SRI's rolle og n\u00f8jagtige mekanisme i forbindelse med PCa stadig ikke fuldt ud forst\u00e5et. Denne unders\u00f8gelse udforsker den molekyl\u00e6re mekanisme for SRI-behandling af PCa gennem in vitro-celleeksperimenter, netv\u00e6rksfarmakologi og molekyl\u00e6re docking-teknikker, hvilket giver et vist forskningsgrundlag og teoretisk grundlag for dets kliniske anvendelse.<br \/>\nH\u00f8j spredningshastighed er en vigtig egenskab ved uendelig v\u00e6kst af maligne tumorceller, og derfor er h\u00e6mning af celleproliferation en effektiv strategi til behandling af kr\u00e6ft. I denne unders\u00f8gelse blev det konstateret, at med stigningen i SRI-koncentrationen blev v\u00e6ksten af DU-145-celler signifikant h\u00e6mmet ved at p\u00e5vise cellelevedygtighed og celleklondannelse, hvilket indikerer, at SRI har evnen til at h\u00e6mme spredningen af DU-145-celler. De prolifererende cellekerneproteiner Ki67 og PCNA er mark\u00f8rproteiner for celleproliferation, der almindeligvis anvendes som pr\u00e6diktive og prognostiske mark\u00f8rer til kr\u00e6ftdiagnose og -behandling. Denne unders\u00f8gelse p\u00e5viste udtrykket af Ki67 i DU-145-celler gennem immunfluorescens og qPCR og fandt, at SRI signifikant kunne h\u00e6mme udtrykket af Ki67. qPCR og Western blot-analyse viste, at SRI signifikant kunne reducere udtrykket af PCNA i DU-145-celler, hvilket yderligere beviste, at SRI har evnen til at h\u00e6mme spredningen af DU-145-celler.<br \/>\nApoptose er hoved\u00e5rsagen til tumorcelled\u00f8d, og derfor kan det at undg\u00e5 apoptose f\u00f8re til resistens i tumorbehandling. Bcl-2 spiller som h\u00e6mmer af apoptose en afg\u00f8rende rolle i reguleringen af celleapoptose. Bax er et homogent konjugat af Bcl-2 og et pro-apoptotisk protein. Forholdet mellem Bcl-2 og Bax er en vigtig indikator for m\u00e5ling af effekten af celleapoptose. Caspase-3 og Caspase-7 er eksekutorer af celleapoptose. N\u00e5r apoptose opst\u00e5r, bliver Caspase-3 og Caspase-7 sekventielt spaltet og aktiveret af opstr\u00f8ms Caspase-proteiner. Derefter vil aktiveret Caspase-3 og Caspase-7 spalte flere downstream-substrater og i sidste ende udl\u00f8se en apoptotisk kaskade-reaktion. Denne unders\u00f8gelse p\u00e5viste apoptose af DU-145-celler efter 48 timers SRI-behandling gennem flowcytometri-eksperimenter, og resultaterne viste, at SRI i v\u00e6sentlig grad kunne fremme apoptose af DU-145-celler. Derfor unders\u00f8gte vi yderligere udtrykket af apoptoserelaterede faktorer. qPCR-resultaterne viste, at SRI kunne opregulere mRNA-ekspressionen af Caspase-3 og Caspase-7, nedregulere mRNA-ekspressionen af Bcl-2 og nedregulere mRNA-ekspressionsniveauet af Bax, men Bcl-2\/Bax-forholdet blev signifikant reduceret (P&lt;0,05). Derudover viste Western blot-resultater, at SRI kunne opregulere ekspressionen af renset-Caspase-3-protein og nedregulere ekspressionen af Bcl-2-protein. Selvom der ikke var nogen signifikant forskel i opreguleringen af Bax-proteinekspressionen, blev proteinekspressionsforholdet mellem Bcl-2\/Bax signifikant reduceret (P&lt;0,05). Disse resultater tyder p\u00e5, at SRI kan regulere udtrykket af apoptoserelaterede faktorer, aktivere apoptosesignalveje og fremkalde apoptose i DU-145-celler.<br \/>\nFor at unders\u00f8ge mekanismen for SRI, der p\u00e5virker spredning og apoptose af DU-145-celler, analyserede denne unders\u00f8gelse det desuden gennem netv\u00e6rksfarmakologiske forskningsmetoder. Vi indsamlede i alt 127 SRI-m\u00e5l gennem HERB, TCMSP, Pharm Mapper og Swiss Target Prediction-databaser og opn\u00e5ede 60 f\u00e6lles m\u00e5l for SRI, der virker p\u00e5 PCa, gennem Venny 2.1.0 online-v\u00e6rkt\u00f8j. Gennem PPI-netv\u00e6rksanalyse identificerede vi 16 kernem\u00e5l (top 10 gradv\u00e6rdier), herunder ALB, TNF, IL6, CASP3, MAPK1, PPARG, MDM2, PGR, MAPK8, NR3C1, ABL1, MAPK14, PARP1, HDAC2, CDK2, GSK3B osv. MAPK8, CASP3, MAPK1, MAPK14 og TNF-kernem\u00e5l er beriget i MAPK-signalvejen. GO-berigelsesanalysen gav 158 poster (P&lt;0,01), hvilket indikerer, at SRI hovedsageligt kan ud\u00f8ve terapeutiske virkninger p\u00e5 PCa ved at regulere genekspression, apoptoseproces, proteinfosforylering og protein serin\/threonin\/tyrosinkinaseaktivitet. Resultaterne af KEGG-vejberigelsesanalysen viste 84 signalveje, herunder kr\u00e6ftsignalvej, proteoglykaner i kr\u00e6ft, MAPK-signalvej, kemokarcinogenese reaktive oxygenarter, PI3K-AKT-signalvej, apoptose osv. (P&lt;0,01). Det mest bekymrende aspekt er MAPK-signalvejens rolle i SRI&#039;s anti-PCA-proces. Mitogenaktiveret proteinkinase (MAPK) kan overf\u00f8re ekstracellul\u00e6r information til kernen og spiller en vigtig rolle i fysiologiske og patologiske processer. Der findes tre typer MAPK-familiemedlemmer i pattedyrceller: ekstracellul\u00e6r signalreguleret kinase (ERK), c-JunN-terminal proteinkinase\/stressaktiveret proteinkinase (JNK\/SAPK) og p38 mitogenaktiveret proteinkinase (p38MAPK, ogs\u00e5 kendt som MAPK14).<br \/>\nP38MAPK er en aktiveret proteinkinase, der deltager i mange biologiske processer som f.eks. celleproliferation, differentiering og apoptose. Dens aktivering kan h\u00e6mme celleproliferation og fremme apoptose. Forskning har vist, at aktivering af p38-stien kan reducere spredningsaktiviteten af BGC-823-mavekr\u00e6ftceller og blokere dem i G2\/M-fasen. Aktivering af p38MAPK kan h\u00e6mme spredningen af SMMC-7721-celler og igangs\u00e6tte den intracellul\u00e6re Caspase-signalkaskade og i sidste ende fremkalde apoptose. Desuden har unders\u00f8gelser vist, at ekstrakter fra traditionel kinesisk medicin kan aktivere p38MAPK, udl\u00f8se Caspase-medierede apoptotiske kaskade-reaktioner og fremkalde apoptose i Lewis-lungekr\u00e6ftceller samt HSC-3- og SCC-9-mundkr\u00e6ftceller. P38MAPK-kinasen spiller ogs\u00e5 en vigtig rolle i forekomsten og udviklingen af PCa. Forskning har vist, at aktivering af p38MAPK kan modst\u00e5 celleproliferation, fremme apoptose og fremkalde tumordvale i ondartet transformation af prostatapitel eller knoglemetastatisk prostatacancer. I mellemtiden har andre unders\u00f8gelser vist, at ingredienser som docetaxel og ursolsyre kan \u00f8ge fosforyleringsniveauet af p38MAPK, fremkalde aldring af PCa-celler, h\u00e6mme spredningen af forskellige PCa-celler som PC-3 og 22Rv1 og fremme deres apoptose. Derfor fokuserer denne unders\u00f8gelse p\u00e5 rollen som p38MAPK-proteinkinase i SRI anti PCa. Ved hj\u00e6lp af molekyl\u00e6r docking-teknologi til at verificere bindingen af SRI til p38MAPK-protein viste resultaterne, at SRI stabilt kan binde sig til p38MAPK-protein med en bindingsenergi p\u00e5 -6,83 kcal\/mol. Detektionsresultaterne af p-p38MAPK og p38MAPK-proteinekspressionsniveauer indikerer, at SRI kan aktivere p38MAPK-protein, og fosforyleringsniveauet af p38MAPK-protein er opreguleret, hvilket tyder p\u00e5, at SRI kan ud\u00f8ve anti-PCA-effekter ved at aktivere p38MAPK-protein og inducere apoptosevej.<br \/>\nSammenfattende kan SRI h\u00e6mme spredningen af DU-145-celler og fremme deres apoptose, hvilket kan v\u00e6re relateret til dets aktivering af p38MAPK. Denne unders\u00f8gelse unders\u00f8gte dog kun effekten af SRI p\u00e5 DU-145-celler in vitro.
