Endnu vigtigere er det, at disse anti-aldringsinterventioner ikke kun forl\u00e6nger levetiden, men ogs\u00e5 forbedrer kroppens generelle sundhed betydeligt. I tidligere unders\u00f8gelser har vi fundet ud af, at EBCQA har potentielle anti-aldringseffekter og kan v\u00e6re relateret til regulering af AKT-aktivitet. I denne unders\u00f8gelse bekr\u00e6ftede vi, at EBCQA har den effekt, at det forl\u00e6nger nematodernes levetid, \u00f8ger oxidations- og varmebestandigheden og forbedrer bev\u00e6geligheden. Gennem mutantstammer af nematoder bekr\u00e6ftede vi, at EBCQA's anti-aldringseffekt er relateret til regulering af PI3K\/Akt\/FoxO3a-vejen, forbedring af antioxidant enzymaktivitet og reduktion af MDA-niveauer; Hos D-galaktose-inducerede aldrende rotter forbedrer EBCQA indl\u00e6rings- og hukommelsesevnen, thymus- og miltkoefficienterne, h\u00e6mmer PI3K- og Akt-aktivering samt FoxO3a-phosphorylering, forbedrer antioxidant-enzymaktiviteten og reducerer MDA-niveauerne.<br \/>\nKomponenter i CQA-klassen er en vigtig klasse af fenoliske aktive ingredienser i Scutellaria baicalensis, og de findes ogs\u00e5 i mange f\u00f8devarer (f.eks. lilla s\u00f8de kartofler, gr\u00f8n kaffe) og l\u00e6geurter. CQA-forbindelser har neurobeskyttende, hjerte-kar-beskyttende, antioxiderende, antiinflammatoriske, antidiabetiske og andre virkninger. Med hensyn til anti-aging forl\u00e6ngede gr\u00f8n kaffeekstrakt og kaprifolieekstrakt, der er rigt p\u00e5 CQA-komponenter, nematodernes levetid, men det er stadig uklart, om deres anti-aging-mekanismer involverer PI3K\/Akt\/FoxO3a-vejen; CQA-komponenternes anti-aging-effekter hos pattedyr er sj\u00e6ldent rapporteret. I overensstemmelse med tidligere rapporter om, at CQA-komponenter har evnen til at forl\u00e6nge nematoders levetid, fandt vi, at EBCQA forl\u00e6nger nematoders levetid med en maksimal forl\u00e6ngelseshastighed p\u00e5 17,7% og forbedrer oxidativ modstand, varmebestandighed og bev\u00e6gelighed. Den D-galaktose-inducerede aldrende rottemodel har vist sig effektivt at simulere naturlig aldring og bruges derfor i vid udstr\u00e6kning til at studere aldringsmekanismer og screene l\u00e6gemidler, der forsinker aldring. Nedsat indl\u00e6rings- og hukommelsesevne er almindelige fysiologiske f\u00e6nomener i forbindelse med aldring, og nogle kan udvikle sig til neurodegenerative sygdomme som Alzheimers sygdom. N\u00e5r alderen stiger, eller der opst\u00e5r sygdomme, falder kroppens immunforsvar som regel gradvist. Thymus og milt er vigtige immunorganer, og deres organindekser kan til en vis grad afspejle st\u00f8rrelsen p\u00e5 kroppens immunforsvar. Derfor er indl\u00e6rings- og hukommelsesevnen samt thymus\/milt-organindekset almindeligt anvendte indikatorer til evaluering af aldring og anti-aldring. D-galaktose-inducerede aldrende rotter viste nedsat indl\u00e6rings- og hukommelsesevne samt nedsat thymus- og miltkoefficient. Indgivelse af EBCQA forbedrede effektivt indl\u00e6rings- og hukommelsesevnen og \u00f8gede thymus- og miltkoefficienterne. Disse resultater indikerer, at EBCQA har en anti-aldringseffekt hos b\u00e5de hvirvell\u00f8se nematoder og hvirveldyr med D-galaktose-induceret aldring.<br \/>\nPI3K\/Akt\/FoxO3a-vejen er en evolution\u00e6rt bevaret signalvej, der reguleres af insulin, IGF-1 og andre faktorer. I reguleringen af aldring spiller PI3K og Akt en rolle i forhold til aldring, mens FOXO3a spiller en rolle i forhold til anti-aldring. I nematoder er AGE-1, AKT-1 og DAF-16 homologer til henholdsvis PI3K, Akt og FoxO3a hos pattedyr. Nematoder med AGE-1-mutationer har en gennemsnitlig levetidsforl\u00e6ngelse p\u00e5 40% til 65%, dem med AKT-1-mutationer har ogs\u00e5 betydeligt l\u00e6ngere levetid, og DAF-16-mutationer forkorter levetiden betydeligt. Hos pattedyr inducerer overekspression af AKT-1 cellul\u00e6r aldring. Mus med AKT-1-mutationer (Akt1+\/-) har en 8% \u00f8get levetid for m\u00e6nd og en 15% \u00f8get levetid for kvinder. Forskning i langlivede befolkninger har ogs\u00e5 vist, at genetiske polymorfismer\/mutationer, der f\u00f8rer til tab af funktion af IGF-1-receptorer, insulinreceptorer eller deres downstream-faktorer (s\u00e5som PI3K, AKT), er forbundet med forl\u00e6nget levetid hos mennesker. Unders\u00f8gelsen af langlivede populationer blandt japanske amerikanere, han-kinesere, californiere, newenglandere, ashkenazi-j\u00f8der, danskere, tyskere og italienere viser, at de genetiske polymorfismer af rs2764264, rs2802292 osv. i FOXO3a er relateret til lang levetid og sund aldring. I reguleringen af PI3K\/Akt\/FoxO3a-stien aktiveres Akt ved phosphorylering af Thr308 og Ser473, mens FoxO3a h\u00e6mmes ved phosphorylering af Thr32 og Ser253. I denne unders\u00f8gelse forl\u00e6ngede EBCQA levetiden for vildtypenematoden N2 og fremmede DAF-16's nukleare translokation, men havde ingen effekt p\u00e5 levetiden for AGE-1-, AKT-1- og DAF-16-mutanter, hvilket indikerer, at EBCQA's rolle i forl\u00e6ngelsen af levetiden afh\u00e6nger af reguleringen af PI3K\/Akt\/FoxO3a-vejen. Rotter behandlet med D-galactose viste opregulering af PI3K-, Akt-, p-Akt- og p-FOXO3a-ekspression, mens FOXO3a-ekspressionen blev nedreguleret. Dette stemmer overens med tidligere rapporter om \u00e6ndringer i D-galactose-inducerede aldrende rotter og naturligt aldrende sm\u00e5\/store rotter. Disse \u00e6ndringer i proteinudtryk kan f\u00f8re til nedsat indl\u00e6rings- og hukommelsesevne samt nedsat thymus- og miltkoefficient og derved fremkalde aldring. Efter indgivelse af EBCQA blev disse \u00e6ndringer h\u00e6mmet, hvilket kan have en anti-aldringseffekt.<br \/>\nI 1956 foreslog den britiske forsker Harmna for f\u00f8rste gang teorien om aldring med frie radikaler og mente, at den cellul\u00e6re dysfunktion for\u00e5rsaget af frie radikalers angreb p\u00e5 biomolekyler er en af de grundl\u00e6ggende \u00e5rsager til aldring i kroppen. I denne unders\u00f8gelse \u00f8gede EBCQA aktiviteten af SOD, GSH Px og CAT i nematoder behandlet med paraquat og rotter behandlet med D-galactose, samtidig med at MDA-niveauet blev reduceret. Den \u00f8gede aktivitet af disse antioxidantenzymer kan f\u00f8re til en reduktion af ROS og oxidativ skade og dermed bidrage til at forsinke aldring. En af de downstream-mekanismer, hvormed PI3K\/Akt\/FoxO3a-vejen p\u00e5virker levetiden, er reguleringen af oxidativ resistens. Hos nematoder udviser nogle langlivede mutantstammer, s\u00e5som daf-2 (e1368) og age-1 (hx546), \u00f8get aktivitet af antioxidantenzymerne SOD, GSH Px og CAT, som er afh\u00e6ngige af DAF-16. Efter DAF-16-mutationen er antioxidant-enzymaktiviteten hos disse mutantstammer n\u00e6sten den samme som hos vildtype-nematoden N2. Hos pattedyr regulerer FoxO3a udtrykket af antioxidantenzymer som SOD, GSH Px, CAT og h\u00e6mmer aktiviteten af FoxO3a, hvilket f\u00f8rer til ophobning af ROS i celler, fremskynder \u00e5reforkalkning og \u00f8ger risikoen for kr\u00e6ft. EBCQA fremmer den nukleare indtr\u00e6ngen af nematoden DAF-16 og h\u00e6mmer fosforyleringen af rotte-FoxO3a, hvilket indikerer, at EBCQA kan spille en anti-aging-rolle ved at \u00f8ge FoxO3a-transkriptionsaktiviteten og antioxidant-enzymaktiviteten.<br \/>\nSammenfattende kan EBCQA effektivt forl\u00e6nge levetiden for nematoder og modvirke D-galactose-induceret aldring hos rotter.
