Ved at h\u00e6mme aktiviteten af \u03b1-amylase for at reducere stivelsesford\u00f8jeligheden kan det forsinke stivelsesford\u00f8jelsen, h\u00e6mme tarmens absorption af glukose, effektivt regulere blodsukkerniveauet i kroppen og dermed lindre symptomerne p\u00e5 diabetes. At tage \u03b1-amylase som terapeutisk m\u00e5l for type 2-diabetes har derfor produceret en r\u00e6kke hypoglyk\u00e6miske l\u00e6gemidler, der anvendes i klinisk praksis, s\u00e5som acarbose, voglilitse, miglitol osv. Disse l\u00e6gemidler har dog alle visse bivirkninger, s\u00e5 der er et presserende behov for at udvikle naturlige hypoglyk\u00e6miske l\u00e6gemidler fra naturlige produkter. For at afklare muligheden for at anvende naturlige produkter som f.eks. polyphenoler fra bipollen til udvikling af naturlige hypoglyk\u00e6miske l\u00e6gemidler er det n\u00f8dvendigt at foretage en grundig unders\u00f8gelse af de h\u00e6mmende virkninger og molekyl\u00e6re mekanismer af polyphenoler fra bipollen p\u00e5 sukkermetabolismeenzymer.<br \/>\nSom en vigtig kornafgr\u00f8de er det blevet rapporteret, at boghvede har hypoglyk\u00e6miske virkninger, og polysaccharider fra boghvedepollen har ogs\u00e5 vist sig at have hypoglyk\u00e6misk aktivitet. Men polyfenoler, som er det vigtigste aktive stof i boghvedebiopollen, er ikke blevet unders\u00f8gt for deres hypoglyk\u00e6miske virkninger. I lyset af dette tager denne unders\u00f8gelse boghvede-bipollen-polyfenoler (BBPP) som forskningsobjekt og unders\u00f8ger dets h\u00e6mmende virkning og mekanisme p\u00e5 alfa-amylase ud fra tre aspekter: enzymkinetik, fluorescensspektroskopi og ultraviolet spektroskopi. Virkningerne af h\u00f8j temperatur, st\u00e6rk alkali, lys og h\u00f8j koncentration af oxidanter og reduktionsmidler p\u00e5 den h\u00e6mmende virkning af BBPP p\u00e5 alfa-amylaseaktivitet blev ogs\u00e5 udforsket. Dette eksperiment kan give et teoretisk grundlag for den omfattende udnyttelse af boghvede-bipollen og udviklingen af naturlige hypoglyk\u00e6miske l\u00e6gemidler og sammensatte formuleringer af alfa-amylaseh\u00e6mmere.<br \/>\nDette eksperiment anvendte forskellige spektroskopiske analyser og h\u00e6mningskinetik til at unders\u00f8ge BBPP's h\u00e6mmende mekanisme p\u00e5 alfa-amylase. Resultaterne viste, at BBPP udviste en st\u00e6rk og reversibel blandet konkurrenceh\u00e6mmende effekt p\u00e5 alfa-amylase, og dens h\u00e6mmende effektivitet var dosisafh\u00e6ngig med en IC50 p\u00e5 2,00 \u00b1 0,06 mg\/ml. Stabilitetsanalysen af BBPP under forskellige forhold viste, at h\u00f8j temperatur, st\u00e6rk alkali, lys og h\u00f8je koncentrationer af oxidanter og reduktionsmidler alle kunne reducere den h\u00e6mmende virkning af BBPP p\u00e5 alfa-amylase. Analyse af slukning af fluorescensspektrum viser, at BBPP udviser statisk slukning ved 273-298K og dynamisk slukning ved 298-310K, med kun \u00e9t bindingssted mellem de to. Synkron fluorescensspektroskopi indikerer, at BBPP for\u00e5rsager foldnings\u00e6ndringer i milj\u00f8et af \u03b1-amylase-tyrosinrester og \u00f8ger hydrofobiciteten. Denne artikel fokuserer p\u00e5 unders\u00f8gelsen af BBPP's h\u00e6mmende effekt p\u00e5 \u03b1-amylase som en hypoglyk\u00e6misk aktiv ingrediens til at lindre postprandial hyperglyk\u00e6mi.
