Den er let at diagnosticere og vanskelig at behandle med en h\u00f8j forekomst og et langt sygdomsforl\u00f8b, hvilket har en negativ indvirkning p\u00e5 patienternes fysiske og mentale sundhed. Psoralea er en t\u00f8rret og moden frugt af b\u00e6lgplanten Psoralea corylifolia L. Det bruges hovedsageligt i traditionel kinesisk medicin til behandling af erektil dysfunktion, natlige emissioner, l\u00e6nde- og kn\u00e6smerter og hv\u00e6sende vejrtr\u00e6kning for\u00e5rsaget af nyre yang-mangel. Det kan ogs\u00e5 bruges lokalt til behandling af vitiligo og alopecia areata. Ikke alene har vores land en historie med at bruge psoriasis til at behandle vitiligo, men der er ogs\u00e5 optegnelser i syd\u00f8stasiatiske lande som Japan, Sydkorea, Indien osv. (kendt som \"Babchi-fr\u00f8\", \"Bakuchi-pulver\", \"Bakuchiol\" osv.). Der findes ogs\u00e5 forskellige doseringsformer til behandling af vitiligo med psoralen, herunder afkog, tinkturer og injektioner (National Medical Standard Z41022361). Selvom der er mange litteraturoptegnelser om behandling af vitiligo med psoralen, er dens virkningsmekanisme og materielle grundlag for behandling af vitiligo ikke fuldt ud forst\u00e5et. Derfor bruger denne artikel ultrah\u00f8jtydende v\u00e6skekromatografi quadrupol time-of-flight massespektrometri (UPLC-Q-TOF-MS) teknologi til at unders\u00f8ge den kemiske sammens\u00e6tning af Psoraleae Fructus alkoholekstrakt (PFE); Ved at udf\u00f8re eksperimenter p\u00e5 zebrafisk for at fremme melaninproduktion blev effekten af PFE p\u00e5 melaninproduktion unders\u00f8gt. Indtil videre er dens \u00e6tiologi og patogenese ikke fuldt ud forst\u00e5et. Forskning har vist, at hudpigmentering er t\u00e6t forbundet med de tre vigtigste metaboliske processer i melanocytter i epidermis, herunder melanocytproliferation, syntese og aktivering af TYR og melaninsyntese, samt vejen for melaninoverf\u00f8rsel fra melanosomer til keratinocytter. Melaninsyntesen er en kompleks reaktion, der foreg\u00e5r i melanocytterne og reguleres af flere faktorer. TYR er det hastighedsbegr\u00e6nsende enzym i melaninsyntesen, og dets aktivitet reguleres af TYRP1 og TYRP2, hvilket g\u00f8r det til en vigtig regulerende faktor i melaninsyntesen.<br \/>\nBaseret p\u00e5 patogenesen og kliniske manifestationer af vitiligo har forskere udviklet en r\u00e6kke dyremodeller i de senere \u00e5r, s\u00e5som kemisk depigmenteringsmiddelinduktion, melanocytstressinduktion, immuninduktion, eksogen T-celleinduktion, transgen, melanocytafl\u00f8sningsinduktion osv. Mus er det vigtigste valg for dyr. If\u00f8lge forskning er 71,4% af humane proteinkodende gener og 82% af patogene gener direkte homologe med zebrafisk, herunder mange gener relateret til humane hudsygdomme. Zebrafiskelarver er gennemsigtige, og den overordnede og cellul\u00e6re struktur i deres hud ligner pattedyrs hud. De har melanocytter og melanosomer, der ligner menneskets, og melanocytterne er nemme at observere under den individuelle udvikling. Derfor er der stadig behov for at udforske dyremodeller for vitiligo yderligere.<br \/>\nPTU er en h\u00e6mmer af TYR, der i vid udstr\u00e6kning bruges til at blokere pigmentering og forbedre den optiske gennemsigtighed i zebrafiskembryoner. Unders\u00f8gelser har vist, at brug af PTU-opl\u00f8sning i stedet for fiskevand efter zebrafiskens tarmembryogenese (5,5 hpf) effektivt kan h\u00e6mme melaninproduktionen og opretholde zebrafiskens gennemsigtighed. Resultaterne af dette eksperiment indikerer, at efter behandling af zebrafisk med PFE i 18 timer efter PTU-modellering, sammenlignet med gruppen behandlet med embryokulturmedium, kan PFE fremskynde regenerering af melanin i \u00f8jne, mave, rygs\u00f8jle og hale af zebrafiskyngel, hvilket er i overensstemmelse med den kliniske anvendelse af psoralen til behandling af vitiligo.<\/p>\n<p><img Baseret p\u00e5 litteratur og praktiske anvendelser valgte denne unders\u00f8gelse derfor 65% ethanol som ekstraktionsreagens. Denne unders\u00f8gelse brugte UPLC-Q-TOF-MS-teknologi til at identificere de kemiske komponenter i PFE. I alt 47 kemiske komponenter blev identificeret, hovedsageligt flavonoider (27) og coumariner (10). De identificerede coumarin-kemiske komponenter PSO og IPSO kan v\u00e6re de vigtigste effektive kemiske komponenter til behandling af vitiligo. Benzofuraner (s\u00e5som psoralen og isopsoralen) kan ogs\u00e5 omdannes til PSO og IPSO i kroppen for at ud\u00f8ve deres virkning. PSO og IPSO er ogs\u00e5 kvalitetskontrolkomponenter for medicinske materialer til knoglefedt, der er specificeret i den kinesiske farmakop\u00e9. Derfor blev PFE og disse to kemiske komponenter valgt til celleeksperimenter. En forel\u00f8big unders\u00f8gelse af virkningsmekanismen viste, at qRT PCR-resultater viste, at sammenlignet med kontrolgruppen \u00f8gede PFE signifikant ekspressionen af Tyr og Tyrp2 mRNA i B16-F10-celler, mens andre behandlingsgrupper kunne \u00f8ge ekspressionen af Tyr, Tyrp1 og Tyrp2 mRNA i varierende grad med statistiske forskelle, hvilket indikerer, at PSO, IPSO og andre potentielt ukendte forbindelser i Fructus Psorale kan ud\u00f8ve terapeutiske virkninger p\u00e5 vitiligo ved at \u00f8ge tyrosinaseindholdet. Der er dog ogs\u00e5 litteraturrapporter om, at PSO virker p\u00e5 den humane melanomcellelinje A375 og h\u00e6mmer aktiviteten af tyrosinasepromotoren; Nedregulering af ekspressionsniveauerne af Tyr, Tyrp1 og Tyrp2 mRNA og h\u00e6mning af melaninproduktionen, i mods\u00e6tning til resultaterne af dette eksperiment, kan skyldes forskellige forskningsemner eller den tovejsregulering af melaninproduktionen af PSO i forskellige koncentrationsomr\u00e5der. Der er behov for yderligere forskning for at kunne drage mere stringente og pr\u00e6cise konklusioner.
