Den er hovedsageligt udbredt i den sydvestlige del af Tianshan-bjergene og den nordvestlige del af Tarim i Xinjiang og er en lokal specialgr\u00f8ntsag, der ofte spises af folk fra alle etniske grupper i Xinjiang. Forskning har vist, at Chamagu-r\u00f8dder indeholder masser af GL'er. Zhuang et al. brugte ultra-high liquid chromatography-teknologi til at analysere typer og indhold af GL'er i forskellige udviklingsstadier og v\u00e6kstdele (r\u00f8dder, st\u00e6ngler, blade) af Xinjiang-roe i l\u00f8bet af deres ern\u00e6ringsm\u00e6ssige v\u00e6kstperiode. Sun har rapporteret, at Qiamagu-fr\u00f8 indeholder fire typer isothiocyanater: phenethylisothiocyanat, tert butylisothiocyanat, isopropylisothiocyanat og allylisothiocyanat.<\/p>\n<p>P\u00e5 nuv\u00e6rende tidspunkt involverer fremstillingen af isothiocyanater hovedsageligt kemisk syntese og enzymatisk hydrolyse. Ved hj\u00e6lp af endogen enzymatisk hydrolyse til fremstilling af isothiocyanater, selvom processen er enkel, kan den endogene myrosinase i planter ikke frigives fuldt ud, hvilket resulterer i lang enzymatisk hydrolysetid og lav effektivitet. Ding sammenlignede m\u00e6ngden af isothiocyanater genereret ved GL-nedbrydning af endogene og eksogene enzymer i rapsfr\u00f8 og fandt, at m\u00e6ngden af isothiocyanater genereret af eksogen myrosinase var 7,6 gange s\u00e5 stor som den endogene myrosinase. GC-MS-identifikation og -analyse viste, at eksogen myrosinase havde en r\u00e6kke forskellige nedbrydningsprodukter, et relativt h\u00f8jt indhold og f\u00e6rre urenheder.<\/p>\n<p>Denne unders\u00f8gelse brugte Chamagu-rod som r\u00e5materiale og anvendte eksogen myrosinase-nedbrydningsmetode til at fremstille Chamagu-isothiocyanat (BRICe). P\u00e5 baggrund af enkeltfaktorscreening blev enzymhydrolysetid, temperatur, pH og ascorbinsyrekoncentration identificeret som v\u00e6sentlige p\u00e5virkningsfaktorer. Et ortogonalt eksperiment med fire faktorer p\u00e5 tre niveauer blev brugt til at optimere ekstraktionsprocessen. Endelig blev den kemiske sammens\u00e6tning analyseret ved hj\u00e6lp af gaskromatografi-massespektrometri (GC-MS), og isothiocyanatets anticancereffekt blev unders\u00f8gt.<\/p>\n<p>Kr\u00e6ft er en af de ondartede sygdomme, der truer menneskers liv. Flere epidemiologiske unders\u00f8gelser har vist, at indtagelse af flere korsblomstrede gr\u00f8ntsager effektivt kan reducere risikoen for kr\u00e6ft. Isothiocyanat er et af produkterne af selv-enzymatisk hydrolyse i korsblomstrede gr\u00f8ntsager. Forskning har vist, at isothiocyanater kan aktivere fase II-enzymer i menneskekroppen, s\u00e5 produkterne fra fase I-reaktionerne kan binde sig til glucuronsyre og glutathion og dermed fremme en hurtig udskillelse af kr\u00e6ftfremkaldende stoffer fra kroppen. Derfor kan isothiocyanater bruges som kr\u00e6fth\u00e6mmer i medicin. Qiamagu er en medicinsk og spiselig plante i Xinjiang, der indeholder forskellige aktive ingredienser som glucosinolater, flavonoider, isothiocyanater og polysaccharider. Men udviklingen og udnyttelsen af Qiamagu-isothiocyanater er i \u00f8jeblikket utilstr\u00e6kkelig. Enzymatisk hydrolyse er i \u00f8jeblikket en milj\u00f8venlig metode til udvinding af isothiocyanater. Ved at optimere betingelserne for enzymatisk hydrolyse kan man ikke kun reducere produktionen af andre sekund\u00e6re metabolitter, men ogs\u00e5 fremskynde produktionen af m\u00e5lproduktet og dermed \u00f8ge udbyttet. Jang fremstillede isothiocyanater ved hj\u00e6lp af endogen myrosinase i Brassicaceae-familien. Selv om den endogene enzymmetode har en enkel proces, kan myrosinasen i planter ikke frigives fuldt ud, hvilket resulterer i lang enzymatisk hydrolysetid og lav effektivitet. Sammenlignet med endogene enzymer er det lettere at kontrollere de enzymatiske hydrolysebetingelser for eksogene enzymer ved at bruge eksogene enzymer til at fremstille isothiocyanater, hvilket er gavnligt for at styre \u00e6ndringer i typerne af enzymatiske hydrolyseprodukter og \u00f8ge m\u00e6ngden af produkter. Derfor er optimering af de eksterne enzymatiske hydrolysebetingelser n\u00f8glen til fremstilling af isothiocyanater.<\/p>\n<p>Dannelsen af enzymatiske hydrolyseprodukter er ikke kun relateret til typen af glucosinolat, men omfatter ogs\u00e5 hydrolysetid, hydrolysetemperatur, pH, tryk og tilstedev\u00e6relsen af Fe2+. Latt \u00e9 fandt ud af, at n\u00e5r opl\u00f8sningens pH er neutral, er det gunstigt for glucosinolater at danne isothiocyanater. Men n\u00e5r pH-v\u00e6rdien er sur\/alkalisk, eller n\u00e5r cykliske svovlkarakteristiske proteiner eller Fe2+ er til stede, vil de enzymatiske hydrolyseprodukter danne en stor m\u00e6ngde nitriler. Yang brugte responsoverflademetoden til at analysere faktorerne pH, EDTA og ascorbinsyre og fandt ud af, at disse tre faktorer har en betydelig indvirkning p\u00e5 ekstraktionsm\u00e6ngden af isothiocyanat. Burmeister fandt ud af, at ascorbinsyre erstattede den katalytiske basefunktion i myrosinases aktive omr\u00e5de og udviste myrosinases katalytiske egenskaber. Tils\u00e6tning af en passende m\u00e6ngde ascorbinsyre til frisk broccoli kan \u00f8ge aktiviteten af myrosinase betydeligt og dermed \u00f8ge produktionen af isothiocyanater. I denne unders\u00f8gelse blev der brugt eksogen myrosinase (fra hvide sennepsfr\u00f8) som nedbrydningsmetode til at fremstille BRIe. De enzymatiske hydrolysebetingelser blev optimeret gennem enkeltfaktor- og ortogonale eksperimenter, og fire faktorer, der havde en betydelig indvirkning p\u00e5 ekstraktionen af BRIe, blev screenet: enzymatisk hydrolysetid, enzymatisk hydrolysetemperatur, ascorbinsyrekoncentration og pH. De optimale enzymatiske hydrolysebetingelser for BRIe blev bestemt som f\u00f8lger: tils\u00e6tning af myrosinase p\u00e5 20 \u03bcL, reaktionstemperatur p\u00e5 70 \u00b0C, reaktionstid p\u00e5 3 timer, pH p\u00e5 6,5, ascorbinsyrekoncentration p\u00e5 0,1 mg\/g og fryset\u00f8rret GL-pulver p\u00e5 2 g. Det er interessant, at den optimale enzymatiske hydrolysetemperatur for denne unders\u00f8gelse er 70 \u00b0C. Der er forskningsrapporter om, at temperaturomr\u00e5det for enzymatisk hydrolyse af planteisothiocyanater fra broccoli er 20-60 \u00b0C, hvor den optimale enzymatiske hydrolysetemperatur er 40 \u00b0C; temperaturomr\u00e5det for udvinding af isothiocyanater fra wasabi-r\u00f8dder er 30-90 \u00b0C, og den optimale enzymatiske hydrolysetemperatur er 70 \u00b0C. Derfor varierer den optimale temperatur for myrosinase afh\u00e6ngigt af oprindelsesarten.<\/p>\n<p>Normalt varierer de dannede typer isothiocyanater afh\u00e6ngigt af betingelserne for enzymatisk hydrolyse. Chen et al. brugte endogen enzymatisk hydrolyse til at identificere de enzymatiske hydrolyseprodukter i broccoli som allylisothiocyanat, isobutylisothiocyanat, 1-butenylisothiocyanat, 4-methylthionitril, 5-methylthiopentonitril, 3-(methylthio)propylisothiocyanat, 5-methylthiopentonitril, 3-(methylthio)propylisothiocyanat, butylisothiocyanat, radise-thiocyanat og radise-sulforafan ved hj\u00e6lp af GC-MS, hvor radise-sulforafan havde det h\u00f8jeste indhold. Yang brugte eksogen enzymatisk hydrolyse til at identificere de enzymatiske hydrolyseprodukter i broccoli som allylisothiocyanat, phenethylisothiocyanat, cyclopentylisothiocyanat, phenylpropanonitril, 2,1-phenyloxazolidinone og skjoldbruskkirtelstimulerende hormon. Denne unders\u00f8gelse analyserede sammens\u00e6tningen af de optimale enzymatiske hydrolysebetingelser for BRIe ved hj\u00e6lp af GC-MS og identificerede syv enzymatiske hydrolyserelaterede produkter, nemlig 3-amino-2-oxazolidinon, 2-oxazolidinethion, 3-buten-1-ylisothiocyanat (BITC), phenylisothiocyanat (PEITC), 1-isothiocyanatpropylester, isothiocyanatisopropylester og 2-phenylisothiocyanat (2-PITC), hvoraf 4 var isothiocyanattyper.<\/p>\n<p>Mange unders\u00f8gelser har vist, at isothiocyanater kan forhindre forekomsten af forskellige kr\u00e6ftformer. Forskning har vist, at PEITC-dosisafh\u00e6ngigt h\u00e6mmer spredningen af musens osteosarkomcellelinje K7M2 og kan for\u00e5rsage G2\/M-cellecyklusstop og apoptose; BITC kan reducere cellelevedygtigheden af gastrisk adenocarcinomcelle AGS betydeligt og fremkalde apoptose og autofagi. Der er dog i \u00f8jeblikket kun f\u00e5 rapporter om isothiocyanaters virkninger og mekanismer p\u00e5 livmoderhalskr\u00e6ft. Denne unders\u00f8gelse fandt gennem en in vitro-cellemodel, at BRIe signifikant h\u00e6mmede spredningen af livmoderhalskr\u00e6ftceller Hela, Siha og U14 p\u00e5 en tids- og dosisafh\u00e6ngig m\u00e5de. Ved at sammenligne 24-timers IC50-v\u00e6rdierne for disse tre celletyper konkluderes det, at Hela-celler er de mest f\u00f8lsomme over for BRIe. Ukontrolleret celleproliferation er den grundl\u00e6ggende \u00e5rsag til kr\u00e6ft, som skyldes forstyrrelse af cellecyklussen. Flowcytometrianalyse afsl\u00f8rede, at BRIe kan standse Hela-cellecyklussen i S- og G2\/M-faserne. Metastasering af kr\u00e6ftceller har altid v\u00e6ret et hotspot for forskningen og en udfordring for den kliniske behandling. Gennem scratch-eksperimenter blev det konstateret, at BRIe effektivt kan h\u00e6mme Hela-cellers migration. Hochest 33258-farvning afsl\u00f8rede, at BRIe kan fremkalde DNA-kondensation i Hela-cellekerner, og der kan observeres synlig granul\u00e6r fluorescens i kernerne, hvilket tyder p\u00e5, at det er et apoptotisk legeme. Yderligere analyse ved hj\u00e6lp af Annexin V-FITC-dobbeltfarvning afsl\u00f8rede, at apoptosehastigheden for Hela-celler behandlet med BRIe steg med stigende koncentration efter 24 timer, hvilket indikerer, at BRIe inducerede apoptose i Hela-celler. Syv enzymatiske hydrolyse-relaterede produkter blev identificeret gennem GC-MS-analyse, og det opn\u00e5ede enzymatiske hydrolyseprodukt BRIe har evnen til at h\u00e6mme spredningen af livmoderhalskr\u00e6ftceller, undertrykke cellemigration, inducere cellecyklusstop og apoptose.
