I mellemtiden betragtes \u00f8rkenmikroalger p\u00e5 grund af deres h\u00f8je olieakkumuleringskapacitet som en lovende ny biobr\u00e6ndstofressource; polysaccharider har p\u00e5 den anden side tiltrukket sig stor opm\u00e6rksomhed i f\u00f8devare-, foder- og medicinalindustrien p\u00e5 grund af deres forskellige funktioner som antikoagulation, antioxidant og antibakteriel. St\u00f8rrelsen af molekylv\u00e6gten, monosaccharidsammens\u00e6tningen og kulstofk\u00e6dens funktionelle grupper er alle relateret til deres biologiske aktivitet.<br \/>\nPolysaccharider klassificeres i sure, neutrale og alkaliske polysaccharider baseret p\u00e5, om de indeholder specifikke funktionelle grupper. De fleste landplanter har dem ikke, fordi sulfatgrupper kan bruges til at modst\u00e5 ekstreme milj\u00f8er som h\u00f8jt saltindhold og h\u00f8j alkali, og indholdet af sulfatgrupper er t\u00e6t forbundet med biologisk aktivitet.<br \/>\nDette eksperiment brugte et centralt kompositdesign (CCD) til at optimere udvindingsprocessen af intracellul\u00e6re polysaccharider (IPS'er) fra \u00f8rkenmikroalger, hvilket kan evaluere eksperimentelle data bedre og mere pr\u00e6cist og l\u00e6gge et godt fundament for yderligere p\u00e5visning af IPS'ers struktur og funktion. Forskning i polysaccharider har altid v\u00e6ret et varmt emne, is\u00e6r i havalger, som er mere popul\u00e6re i offentligheden med hensyn til funktionalitet og sikkerhed. Der er dog relativt lidt forskning i polysaccharider i \u00f8rkenmikroalger.<\/p>\n<p>\u00d8rken-Chlorella har en st\u00e6rk modstandsdygtighed over for salt, alkali og t\u00f8rke, men der er relativt lidt forskning i udvinding af polysaccharider og p\u00e5visning af antioxidantaktivitet. Denne artikel optimerer udvindingsprocessen af IPS'er fra \u00f8rkenmikroalger og udf\u00f8rer separation, rensning og aktivitetsdetektion. CCD-metoden blev brugt til at optimere IPS-ekstraktionsprocessen, og den h\u00f8jeste IPS-ekstraktionsm\u00e6ngde blev opn\u00e5et under betingelser med en temperatur p\u00e5 80 \u00b0C, en tid p\u00e5 1 time og et forhold mellem fast stof og v\u00e6ske (g\/mL) p\u00e5 1:25. To sure polysaccharider, IPS-2 og IPS-3, blev med succes adskilt ved hj\u00e6lp af ionkromatografi med et sulfatindhold p\u00e5 henholdsvis (5,69 \u00b1 0,11)% og (8,46 \u00b1 0,03)%. I mellemtiden best\u00e5r IPS-, IPS-2- og IPS-3-pr\u00f8ver hovedsageligt af glukose og galaktose. IPS fra \u00f8rkenmikroalger viste en h\u00e6mmende effekt p\u00e5 DP-PH-frie radikaler med en IC50 p\u00e5 0,011 mg\/ml, hvilket er t\u00e6t p\u00e5 IC50 for VC p\u00e5 DPPH-frie radikaler p\u00e5 0,007 mg\/ml; med hensyn til hydroxylradikaler viste IPS fra \u00f8rkenmikroalger en h\u00e6mmende effekt med en IC50 p\u00e5 0,737 mg\/ml, hvilket ogs\u00e5 er t\u00e6t p\u00e5 IC50 p\u00e5 0,626 mg\/ml for VC p\u00e5 hydroxylradikaler. Denne ekstremt st\u00e6rke antioxidantaktivitet kan v\u00e6re relateret til indholdet af monosakkarid (glukose). Forskningsresultaterne i denne artikel giver et litter\u00e6rt grundlag for strukturen og aktiviteten af polysaccharider fra \u00f8rkenmikroalger og l\u00e6gger et akademisk fundament for udviklingen af potentielle produkter inden for flere omr\u00e5der s\u00e5som f\u00f8devarer, medicin, kosmetik osv.
