Study on the Chemical Composition of the Ethyl Acetate Part of Black faced God
Breynia fruticosa is a perennial shrub of the Euphorbiaceae family, with tender branches and leaves. Distributed in provinces and regions such as Zhejiang, Fujian, Guangdong, Hainan, Guangxi, Sichuan, Guizhou, Yunnan, etc., it is scattered in shrubs or forest edges on mountain slopes, flatlands, and open fields. It is also known as ghost painting talisman, pot cover wood, green pill wood, dog foot thorn, etc. It has the effects of clearing heat and dampness, promoting blood circulation and detoxification. Can be used to treat gastroenteritis, sore throat, rheumatic bone pain, eczema, hyperlipidemia, rheumatic rheumatism, postpartum milk obstruction, itching, etc; Boiling the whole plant in water and washing it externally can treat sores, boils, dermatitis, etc. Previous research has shown that the main chemical components of blackface include flavonoids, lignans, coumarins, sesquiterpenes, triterpenes, protocatechuic aldehydes, steroids, polysaccharides, organic acids, etc. Pharmacological studies have shown that Heimian Shen has anti-inflammatory, antibacterial, antiviral, immunosuppressive effects, anti skin type I hypersensitivity effects, tyrosinase inhibition effects, and anti mouse chronic dermatitis and eczema effects. Based on the research conducted by the research group on the antiviral effects of heat clearing and detoxifying plants, it was found that the 95% ethanol extract of Black Face God has significant in vitro inhibitory activity on EB virus DNA fragmentation and replication. Therefore, in order to search for active ingredients against EBV virus, we used activity oriented separation method and found that the active ingredients were present in the ethyl acetate extraction site. We systematically studied the chemical composition of this component in order to clarify the pharmacological substance basis of Black faced God against EBV and provide theoretical basis for further development and utilization of its medicinal value.

 

Black faced god, as a perennial shrub used for medicinal purposes, is widely distributed in southern China and has abundant plant resources. In this experiment, modern chromatographic techniques were used to study the chemical composition of the ethyl acetate fraction of the 95% extract of Black faced God. 24 compounds were isolated from the extract, including compounds 1-11, lignin compounds, compounds 15 and 16, tigenin compounds, compounds 17-22, triterpenoids and steroids, and other compounds such as aromatic alcohols, esters, and alkaloids. Except for compounds 1, 3, 4, 8, and 22, all of the above compounds were obtained from this plant for the first time. In addition, the obtained compounds were screened for their inhibitory activity on EBV lysis and replication, as well as their cytotoxic activity. From the results, compounds 12 and 14 showed a more significant inhibitory effect on EBV lysis and replication, with compound 14 exhibiting higher EBV inhibition activity than dextrin. According to literature reports, current studies on the antiviral active ingredients of Black Mask God mainly focus on influenza virus, vesicular stomatitis virus gland, and virus. To further investigate the material basis of antiviral activity, we will continue to study its chemical composition, with a particular focus on alkaloids, in order to obtain compounds with good inhibitory activity on EBV cleavage and replication. This study enriches the chemical composition information of the black faced god and provides a material basis and scientific basis for further developing its medicinal value to a certain extent.
