Research on Metabolites of Pseudomonas Ovale, an Endophytic Bacteria in Chuanxiong
During the process of mutualistic symbiosis with the host, endophytic bacteria in plants also produce many metabolites with pharmacological activity. Chuanxiong is the dried rhizome of Ligusticum chuanxiong Hort., a plant in the Umbelliferae family. It has a pungent and slightly bitter taste, and a warm nature. It has the effects of promoting blood circulation, dispelling wind and relieving pain. At present, there is a large amount of research on the chemical composition and pharmacological activity of Ligusticum chuanxiong, but there is still relatively little research on the chemical composition of endophytic bacteria in Ligusticum chuanxiong. There are various types of endophytic fungi in Ligusticum chuanxiong, and Wang et al. isolated 87 endophytic fungi from the rhizomes of Ligusticum chuanxiong; Scholars have studied the metabolites of the endophytic bacteria Cladosporium sp. and Fusarium tricintum in Chuanxiong, and isolated some structurally novel and highly active compounds from them. The Pseudomonas genus contains 10 bacterial species, namely P bakeri、P. irregulare、P. jaipurense、P. desertorum、P. luteolum、P. ovale、P. punctatum、P. zonatum、P. macroglobosum、P. hygrophilum。 From the late 1960s to the early 21st century, some scholars conducted a series of studies on the metabolites of Pseudomonas aeruginosa and isolated phospholipids, pseurotins A-E, and cell relaxins G, X, Y, and Z from them. The research group had been engaged in the study of the chemical composition of endophytic bacteria in traditional Chinese medicine, and obtained a strain of P. from the root of Chuanxiong Ovale endophytic bacteria were subjected to large-scale fermentation using PDB medium in this study. The secondary metabolites were isolated, purified, and structurally identified, and their anti-inflammatory activity was detected in order to identify compounds with pharmacological activity and enrich the chemical composition research of Ligusticum chuanxiong endophytic bacteria.

This study isolated an endophytic bacterium P. from Ligusticum chuanxiong ovale, A new polyketone compound and four known compounds were isolated from the ethyl acetate extract of the expanded fermentation by macroporous resin, medium pressure C18, medium pressure MCI and gel column. All five compounds were isolated for the first time from this genus of bacteria. This time P The metabolic species separated by ovale fermentation were analyzed by high-performance liquid chromatography, which showed that there were still many compounds, mostly homologous, in the methanol fraction of D101 macroporous resin after 60% (Fr.3). However, due to insufficient fermentation, they were not isolated. Therefore, it is speculated that there are still many undiscovered chemical components in this bacterium, including compounds with novel structures worth exploring; Due to the good anti-inflammatory activity of compound 1, according to literature reports, compound 2 also has strong anti-inflammatory activity. In addition to the above activities, compound 3 shows moderate antibacterial activity against MRSA, with a MIC value of 80 μ g/m; Compound 4 exhibits cytotoxic activity against P388 and A-549 cell lines, and has a significant inhibitory effect on liver cancer cells (CBRH-7919), such as P Further research by ovale is expected to yield more compounds with good pharmacological activity.
